Generaci\u00f3n de los ratones deficientes en dna polimerasa m.  la dna polimerasa mu (polm) es una dna polimerasa perteneciente a la familia x de dna polimerasas. Debido a su expresi\u00f3n en \u00f3rganos linfoides secundarios, a su identidad con la transferasa terminal (42%) y a su capacidad mutag\u00e9nica, se propuso como uno de los agentes mutadores en el proceso de hipermutaci\u00f3n som\u00e1tica de los genes de las inmunoglobulinas. Para determinar el papel espec\u00edfico de polm en este proceso se obtuvo y secuenci\u00f3 ellocus gen\u00f3mico murino de esta polimerasa y generamos el modelo knockout correspondiente. los ratones deficientes en polm son viables y f\u00e9rtiles, sin anormalidades aparentes.  2. Polm es uno de los componentes del mecanismo de hipermutaci\u00f3n som\u00e1tica.  el an\u00e1lisis del proceso de hipermutaci\u00f3n som\u00e1tica en los animales deficientes en poim revel\u00f3 que las c\u00e9lulas b son capaces de hipermutar y generar una respuesta id\u00e9ntica a la de los controles. Sin embargo, las c\u00e9lulas que est\u00e1n hipermutando (los centroblastos) se acumulan en los \u00f3rganos linfoides secundarios de los ratones knockout. El an\u00e1lisis del proceso de hipermutaci\u00f3n mostr\u00f3 una reducci\u00f3n en el n\u00famero de clones altamente mutados (con m\u00e1s de 10 mutaciones). La hipermutaci\u00f3n tiene lugar durante la reacci\u00f3n del centro germinal, durante la cual el centroblasto sufre m\u00faltiples rondas de hipermutaci\u00f3n hasta que adquiere suficientes mutaciones como para incrementar su afinidad por ant\u00edgeno y escapar dela reacci\u00f3n. proponemos que la deficiencia en polm reduce la tasa de hipermutaci\u00f3n y el centroblasto requiere m\u00e1s rondas de hipermutaci\u00f3n para escapar de la reacci\u00f3n del centro germinal, lo que explica la acumulaci\u00f3n de centroblastos.  3.
