Putida u. Este locus tiene una extensi\u00f3n de 7.000 pb y contiene 6 genes (phac1, phaz, phac2, phad, phaf y phai). Se identificaron tres regiones de dna con actividad promotora. Estos promotores gobiernan la expresi\u00f3n de los genes de este locus. La deleci\u00f3n completa del locus pha en p. Putida u nos permiti\u00f3 obtener un mutante incapacitado para acumular pol\u00edmero tanto a partir de precursores alif\u00e1ticos como arom\u00e1ticos. En este mutante, la expresi\u00f3n tn trans del gen phac1 (que codifica la actividad polimerasa) da lugar a la acumulaci\u00f3n intracelular de pol\u00edmero. No obstante, la acumulaci\u00f3n tiene lugar siguiendo un patr\u00f3n (caracterizado por la aparici\u00f3n de un elevado n\u00famero de gr\u00e1nulos de peque\u00f1o tama\u00f1o) claramente diferenciado del de la cepa silvestre. Este hecho nos indica que, si bien es posible la acumulaci\u00f3n de pol\u00edmero \u00fanicamente con la expresi\u00f3n del gen phac1, ser\u00eda necesario el concurso de otros factores para que la acumulaci\u00f3n tenga lugar seg\u00fan el modelo propio de la cepa silvestre (pocos gr\u00e1nulos de gran tama\u00f1o).  sobre la cepa delecionada en el locus pha en la que se expresa el gen phac1, se clonaron los genes phad, phaf, phai y combinaciones de ellos, utilizando otro pl\u00e1smido compatible. La expresi\u00f3n de phaf (solo o en combinaci\u00f3n con phad) da lugar a la aparici\u00f3n de unos gr\u00e1nulos similares en n\u00famero y tama\u00f1o a los de la cepa silvestre. Este hecho sugiere que el producto del gen phaf desempe\u00f1a un papel fundamental en el establecimiento de la arquitectura molecular del gr\u00e1nulo; posiblemente dirigiendo y\/o controlando los mecanismos que desencadenan la coalescencia de los gr\u00e1nulos incipientes para generar el gr\u00e1nulo maduro, con la consiguiente disposici\u00f3n y estabilizaci\u00f3n intracelular del material polim\u00e9rico.
