Se desarrollaron 3 cepas bcg recombinantes expresando la prote\u00edna completa gp120 de la envuelta del vih, cepa (hxbc2). Se evaluaron diferentes promotores de bcg para inducir la expresi\u00f3n de la prote\u00edna heter\u00f3loga del vih. Inicialmente, usamos el promotor hsp60 de bcg, un promotor fuerte que ha demostrado ser uno de los m\u00e1s eficaces y posteriormente, se prob\u00f3 con un promotor d\u00e9bil denominado- ant\u00edgeno. Una vez obtenidas las diferentes cepas bcg recombinantes se evalu\u00f3 la expresi\u00f3n in vitro de la prote\u00edna mediante la t\u00e9cnica de western blot.  posteriormente evaluamos in vivo la respuesta inmune celular espec\u00edfica, en un modelo animal murino. Se inmunizaron ratones con las cepas bcgr: 222viha, bcgr: 223viha y virus vaccinia modificado cepa ankara (vam), expresando el inmunogeno del viha correspondiente a la prote\u00edna entera gag y envuelta del subtipo a predominante en africa del este, m\u00e1s diferentes etp\u00edtopos ctl de nef, pol. La respuesta espec\u00edfica de c\u00e9lulas t cd8+, se detect\u00f3 por la tinci\u00f3n de los tetr\u00e1meros y por la tinci\u00f3n intracelular de ifn-. finalmente para superar las limitantes en la diferenciaci\u00f3n de la inducci\u00f3n de citocinas entre bcg (cepa salvaje) y bcgr por la t\u00e9cnica de elispot, evaluamos la expresi\u00f3n g\u00e9ncia de ifn-. A partir de linfocitos de rat\u00f3n, delectando el arnm de esta citocina por la t\u00e9cnica de rt-pcr.  aportaciones de la tesis:  * dise\u00f1o y desarrollo, por ingenier\u00eda gen\u00e9tica y biolog\u00eda molecular de 3 cepas vacunales de bcg recombinantes.  bcgr: 261 vih-1gp120  bcgr: 222 vih-1gp120  bcgr: 223 vih-1gp120  * estudio y descripci\u00f3n del reajuste gen\u00e9tico que produce cepa salvaje, cuando esta es transformada por el vector de expresi\u00f3n micobacteriano pmv261 que incluye conado el fragmento de adn que codifica la prote\u00edna del vih-1
