Se pueden integrar y expresar varios genes en forma policistr\u00f3nica bajo el control de un \u00fanico promotor, ya que los mecanismos de expresi\u00f3n son similares a los bacterianos. Una ventaja medioambiental importante es que se evita la dispersi\u00f3n del transg\u00e9n a otras plantas, debido a la ausencia de plastidios en los granos de polen. Adem\u00e1s no se ha descrito el silenciamiento g\u00e9nico ni existe el denominado efecto de posici\u00f3n debido al lugar de inserci\u00f3n del transg\u00e9n puesto que la integraci\u00f3n se realiza de forma dirigida por recombinaci\u00f3n hom\u00f3loga este \u00faltimo aspecto disminuye mucho el volumen de trabajo a la hora de seleccionar las l\u00edneas transformadas.El presente proyecto tiene como objetivos analizar la viabilidad de la transformaci\u00f3n plastidial para la producci\u00f3n de ant\u00edgenos vacunales y comprobar la respuesta inmune desarrollada tras su administraci\u00f3n a animales de laboratorio. Para ello se expres\u00f3 en cloroplastos de tabaco el p\u00e9ptido 2l21, que confiere protecci\u00f3n en perros frente al parvovirus canino (cpv), fusionado al extremo carboxi-terminal de la subunidad b de la toxina del c\u00f3lera (ctb) o de la prote\u00edna de fluorescencia verde (gfp). Se obtuvieron unos altos niveles de expresi\u00f3n de las prote\u00ednas de fusi\u00f3n (hasta el 31% respecto a la prote\u00edna soluble total). Una \u00fanica planta es capaz de producir 310 mg del ant\u00edgeno vacunal. Las dos prote\u00ednas de fusi\u00f3n fueron estables tanto en hojas j\u00f3venes como viejas y la vida media estimada, determinada por marcaje y pulso-caza, fue superior a 48 h en ambos casos.
