Se ha surgido, pues, que el ngf podr\u00eda promover sprouting en la zona de la lesi\u00f3n y generar cambios pl\u00e1sticos en la primera neurona sensorial y sus proyecciones centrales, colaborando de este modo al inicio del dolor neurop\u00e1tico.  el objetivo general de esta investigaci\u00f3n fue evaluar estos posibles mecanismos. para realizar el estudio se utiliz\u00f3 un abordaje metodol\u00f3gico directo: la administraci\u00f3n endoneural del ngf o veh\u00edculo. Se utilizaron ratas sprague-dawley macho de 250 a 300 g, cuyos nervios ci\u00e1ticos izquierdos fueron inyectados con 10 ul de soluci\u00f3n. Se realizaron pruebas conductuales de dolor neurop\u00e1tico est\u00edmulo-dependiente (hiperlagesia t\u00e9rmica y alodinia mec\u00e1nica), de dolor inflamatorio (prueba de la formalina), se realiz\u00f3 un estudio histol\u00f3gico del nervio ci\u00e1tico inyectado y un estudio inmunohistoqu\u00edmico para los p\u00e9ptidos sensoriales sp y p\u00e9ptido relacionado al gen de la calcitonina (calcitonin gene-related peptide, cgrp) en el ganglio de la ra\u00edz dorsal (dorsal root ganglion, drg) y el asta posterior de la m\u00e9dula espinal. La administraci\u00f3n endoneural de ngf (1 a 30 ng) produjo hiperalgesia t\u00e9rmica, pero no alodinia mec\u00e1nica. Adem\u00e1s, el ngf produjo desmielinizaci\u00f3n, una modesta degeneraci\u00f3n de fibras e indujo la formaci\u00f3n de brotes axonales. Estos efectos conductuales e histol\u00f3gicos del ngf fueron comparables y vinculables en cuanto a su relaci\u00f3n dosis-efecto como en cuanto a su curso temporal, lo que sugiere que ambos efectos pueden estar relacionados. La administraci\u00f3n endoneural de 30ng del ngf produjo un aumento significativo tanto del porcentaje de c\u00e9lulas medianas a grandes que contienen cgrp en el drg, como del \u00e1rea ocupada por
