El n\u00facleo est\u00e1 rodeado por una doble membrana, la envuelta nuclear, que es capaz de acumular calcio en el interior de su lumen. Se ha propuesto que el calcio puede ser liberado directamente desde la envuelta nuclear hacia el citoplasma a trav\u00e9s de canales i\u00f3nicos asociados tanto a receptores de inositol trisfosfato presentes en la membrana interna de la envuelta nuclear. Por otro lado, el citosol y el nucleoplasma est\u00e1n comunicados a trav\u00e9s de poros nucleares de 10 nm de di\u00e1metro, aparentemente susceptibles al paso libre de iones. En primer lugar se miden los niveles de calcio en el interior de las mitocondrias de c\u00e9lulas gh3 vivas mediante im\u00e1genes de bioluminiscencia obtenidas a partir de la sonda de calcio aequorina (aeq) dirigida a las diferentes organelas. Para medir la [ca2+], se expresa la aeq en c\u00e9lulas gh3 mediante la infecci\u00f3n con un vector v\u00edrico del tipo herpes simplex (hsv). As\u00ed se comprueba que existen oscilaciones de calcio en las mitocondrias que se comportan de forma similar a las citos\u00f3licas. el significado fisiol\u00f3gico de estas oscilaciones de calcio mitocondrial podr\u00eda ser la modulaci\u00f3n de la respiraci\u00f3n. El incremento de la [ca2+]m a niveles de micromolar es capaz de activar numerosas deshidrogenasas mitocondriales que resulta en un aumento de la respiraci\u00f3n. Otro de los objetivos de esta tesis doctoral es comparar las se\u00f1ales de calcio citos\u00f3licas y nucleares registradas mediante aeqs dirigidas tanto al citosol como al n\u00facleo de diversos tipos celulares, tanto en reposo como frente a distintos est\u00edmulos. para comprobar si la envuelta nuclear afecta a la progresi\u00f3n de la onda de calcio se inducen incrementos de la [ca2+]c tanto por activaci\u00f3n de la entrada de calcio a trav\u00e9s de la membrana plasm\u00e1tica por despolarizaci<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
