Nosotros utilizamos ratones knockout para el cftr (cepa cftrtm1cam, normales y -\/- (cf)) en el estudio de los mecanismos de secreci\u00f3n hidroelectrol\u00edtica en conductos pancre\u00e1ticos aislados. Los objetivos que nos planteamos en este trabajo fue analizar la secreci\u00f3n de fluido estimulada por secretagogos que act\u00faan a trav\u00e9s de la ruta del ampc (secretina y forskolina) y del calcio (carbacol, agonista colin\u00e9rgico de receptores muscar\u00ednicos) en conductos pancre\u00e1ticos de ratones cf, as\u00ed como investigar los mecanismos responsables de esta secreci\u00f3n de fluido (ambas cosas en comparaci\u00f3n con ratones normales) y si esta secreci\u00f3n se deb\u00eda a fen\u00f3menos de transporte epitelial de cl- y\/o de hco3-. Realizamos estudios funcionales, en conductos pancre\u00e1ticos aislados de rat\u00f3n, cf y normales, mediante t\u00e9cnicas de videomicroscop\u00eda digital, y de registro de ph y calcio intracelular mediante microfluorimetr\u00eda. nuestros resultados nos permiten concluir: los conductos pancre\u00e1ticos del rat\u00f3n secretan fluido en respuesta a la estimulaci\u00f3n colin\u00e9rgica, secreci\u00f3n mediada por el transporte de cl- y hco3-; las c\u00e9lulas ductulares pancre\u00e1ticas de ratones cf secretan fluido significativamente en respuesta a secretina y forskolina , secreci\u00f3n mediada por el transporte de cl- y de hco3-; como en ratones normales, la secreci\u00f3n de cl- en c\u00e9lulas ductulares pancre\u00e1tica cf est\u00e1 mediada por la actividad de un cotransportador basolateral na+ k+ 2cl-, y la de hco3- por la actividad del intercambiador na+\/h+, interviniendo adem\u00e1s un cotransportador na+ hco3-; los canales de cl- dependientes de ca2+ (cacc) no son responsables de la secreci\u00f3n estimulada por forskolina en c\u00e9lulas ductulares de ratones cf; en la membrana luminal de estas c\u00e9lulas existe un
