En nuestro trabajo, estudiamos el efecto de elastasa y catepsina g, dos proteasas presentes en los granulos primarios de los neutrofilos, sobre moleculas componentes de las membranas basales nativas, en concreto las membranas basales de cristalino y glomerulo renal bovinos, y el posible contenido informacional de los fragmentos solubilizados.  ambas proteasas son capaces de degradar las membranas basales de cristalino y glomerulo bovino (mbc y mbg) y solubilizar colageno iv, laminina y fibronectina.  elastasa es mas efectiva en la solubilizacion de proteinas, y en particular de colageno iv. En mbc el substrato mayoritario de elastasa es el colageno iv que representa casi el 100% del material solubilizado, sin embargo solo un 25% del material degradado por catepsina g pertenece a esta molecula. La solubilizacion de colageno iv a partir de mbg por elastasa y catepsina g corresponde a un 31 y 7% de la proteina solubilizada respectivamente. No existe un efecto cooperativo en la degradacion de la mb cuando estas dos proteasas actuan simultaneamente. La principal region del colageno iv susceptible de hidrolisis es la region triple helicoidal, particularmente la de la cadena alfa 1(iv). El dominio nc1 de la molecula de colageno iv en estado nativo es resistente a elastasa y catepsina g, sin embargo sus subunidades disociadas son sensibles a la accion enzimatica. El material solubilizado por elastasa a partir de mbc puede influir en los procesos que se suceden en la activacion de los neutrofilos. Dichos fragmentos pueden actuar como agonistas quimiotacticos aumentando la motilidad de los leucocitos al doble que en ausencia de estimulo.
