{"id":76011,"date":"2018-03-09T23:21:19","date_gmt":"2018-03-09T23:21:19","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/modulacion-del-desarrollo-y-la-supervivencia-de-las-neuronas-granulares-de-cerebelo-por-el-nmda-mecanismos-moleculares-implicados\/"},"modified":"2018-03-09T23:21:19","modified_gmt":"2018-03-09T23:21:19","slug":"modulacion-del-desarrollo-y-la-supervivencia-de-las-neuronas-granulares-de-cerebelo-por-el-nmda-mecanismos-moleculares-implicados","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/modulacion-del-desarrollo-y-la-supervivencia-de-las-neuronas-granulares-de-cerebelo-por-el-nmda-mecanismos-moleculares-implicados\/","title":{"rendered":"Modulaci\u00f3n del desarrollo y la supervivencia de las neuronas granulares de cerebelo por el nmda: mecanismos moleculares implicados"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Francesc Xavier Xifr\u00f3 Collsamata <\/strong><\/h2>\n<p>Las neuronas granulares de cerebelo (cgcs) necesitan la aportaci\u00f3n continua de estimulaciones excitadoras para sobrevivir durante el proceso postnatal de migraci\u00f3n desde la capa granular externa hasta la capa granular interna. si las cgcs no reciben estas estimulaciones, mueren por apoptosis.  en cultivo, las cgcs mueren por apoptosis en presencia de concentraciones fisiol\u00f3gicas de potasio (5mm; k5) pero sobreviven en presencia de concentraciones desporalizantes de potasio (25mm; k25) o del agonista glutamat\u00e9rgico n-metil-d-asp\u00e1rtico (nmda). Con estos antecedentes, nosotros observamos que la adici\u00f3n del nmda o de k25 durante solo 24 horas en las cgcs inmaduras (a 2 d\u00edas in vitro; div) promueve la supervivencia de las cgcs a lo largo de los divs (hasta 8 div). Este efecto neuroprotector se producir\u00eda, al menos en parte, a trav\u00e9s de una inhibici\u00f3n de la caspasa-3 (una proteasa clave en el desencadenamiento de la apoptosis). Utilizando inhibidores farmacol\u00f3gicos, observamos que la inhibici\u00f3n de la caspasa-3  por el nmda es dependiente de la v\u00eda pi3k\/akt y de las tirosinas quinasas; mientras que la inhibici\u00f3n por k25 seria dependiente de la pi3k\/akt y de la camkii. Parte de la inhibici\u00f3n de la caspasa-3 por nmda o k25 se producir\u00eda por una inhibici\u00f3n de la v\u00eda intr\u00ednseca de la apoptosis, tambi\u00e9n llamada v\u00eda mitocondrial. Nmda o k25 son capaces de inhibir diferentes elementos proapopt\u00f3ticos relacionados con la v\u00eda mitocondrial, como son: disminuci\u00f3n del potencial transmembrana mitocondrial, activaci\u00f3n de prote\u00ednas propapot\u00f3ticas de la familia bcl-2 y liberaci\u00f3n de prote\u00ednas proapopt\u00f3ticas mitocondriales. Adem\u00e1s, son capaces de activar prote\u00ednas antiapopt\u00f3ticas de la familia bcl-2. El conjunto de estos procesos contribuir\u00eda en la inhibici\u00f3n de la caspasa-3 que observamos en presencia de nmda o k25. Observamos tambi\u00e9n que, en k25, la v\u00eda mitocondrial seria regulada, al menos en parte, por la v\u00eda jnk.
