Las adp-ribosa hidrolasas son potenciales agentes protectores que impiden la acumulaci\u00f3n intracelular de adp-ribosa. En mam\u00edferos, se conocen dos tipos de enzimas con actividad adp-ribosa hidrolasa:     1,- adp-ribosa pirofosfatasas altamente espec\u00edficas de baja km (menor 1 um) que de manera dependiente de mg2+ hidrolizan s\u00f3lo adp-ribosa y el an\u00e1logo no fisiol\u00f3gico idp-ribosa.     2,- nucle\u00f3sido difosfoaz\u00facar o difosfoalcohol (ndp-x) pirofosfatasas menos espec\u00edficas, que adem\u00e1s de a(i)dp-ribosa hidrolizan otros sustratos ndp-x no reductores.     en esta tesis, describimos que extractos de placenta humana contienen dos adp-ribosa hidrolasas, que fueron caracterizadas tras una purificaci\u00f3n de unas 1000 veces. Una de ellas, es una adp-az\u00facar pirofosfatasa: hidroliz\u00f3 adp-ribosa, adp-glucosa y adp-manosa, pero no por ejemplo udp-glucosa, a velocidades similares. Se parece a una enzima previamente estudiada en eritrocitos humanos y a la expresada a partir de clones denudt5 humano, pero presenta una km para adp-ribosa 5-20 veces menor (7 um). la otra es una adp-ribosa pirofosfatasa altamente espec\u00edfica de baja km (0,4 um), siendo la primera enzima de estas caracter\u00edsticas identificada en tejidos humanos. La enzima recombinante humana nudt9, estudiada tras su expresi\u00f3n en e.Coli, present\u00f3 propiedades similares a las de la adp-ribosa pirofosfatasa de placenta, incluyendo el bajo valor de km, lo que nos permiti\u00f3 establecer la identidad de ambas enzimas. Al ser incubadas conh2o2, tanto la adp-ribosa pirofosfatasa purificada de placenta como nudt9 presentaron una parente \u00abcambio de especificidad de cati\u00f3n\u00bb.
