La unidad b\u00e1sica de la cromatina es el nucleosoma, que consiste en 147 pb de dna ensamblados alrededor de un oct\u00e1mero de histonas, formado por dos copias de cada una de las histonas, h2a, h2b, h3 y h4, cada histona contiene dos motivos funcionales, el motivo histone fold que participa en interacciones histona-histona y tambi\u00e9n en interacciones histona-dna, y las colas de los extremos nh2-terminal, que es susceptible de sufrir modificaciones postraduccionales, tales como acetilaci\u00f3n, metilaci\u00f3n, fosforilaci\u00f3n, ubiquitinaci\u00f3n y sumoilaci\u00f3n. Estas colas tambi\u00e9n participan en la interacci\u00f3n con nucleosomas adyacentes. Adem\u00e1s de las histonas internas, la cromatina de eucariotas tambi\u00e9n contiene la histona h1, que se dispone en el exterior del nucleosoma, y otras prote\u00ednas no histona como las hmgs (high mobility group). La acetilaci\u00f3n de histonas es una de las modificaciones postraduccionales que m\u00e1s se ha estudiado. Esta modificaci\u00f3n es realizada por complejos hlstona acetiltransferasa (ha ts), y es revertida por complejos histona desacetilasa (hds). En base a la localizaci\u00f3n subcelular, la preferencia de histona y la habilidad de modificar histonas nucleosomales, los complejos ha t nativos se han clasificado en dos grupos. Los enzimas hat tipo a son enzimas nucleares capaces de acetilar hlstonas ensambladas en cromatina, y que frecuentemente se asocian con procesos de regulaci\u00f3n de la transcripci\u00f3n. Los enzimas ha t tipo b tienen una localizaci\u00f3n principalmente citoplasm\u00e1tica y \u00fanicamente acetilan histonas libres, presumiblemente antes de su deposici\u00f3n en el dna para formar nucleosomas. En levadura se describi\u00f3 la presencia de una hat tipo b, [ruiz-garc\u00eda et al., 1998], que se aisl\u00f3 en la fracci\u00f3n citopl\u00e1smlca y que tenia una masa molecular de 200 kda.
