En este trabajo se ha estudiado el mecanismo molecular de regulaci\u00f3n de la expresi\u00f3n g\u00e9nica mediada por la  ruta pkc, tomando como modelo en gen fks1 que codifica para la subunidad catal\u00edtica de la glucano sintasa. Se ha  caracterizado que la secuencia m\u00ednima responsable de la regulaci\u00f3n mediada por la ruta pkc en el gen fks1  corresponde con una secuencia de uni\u00f3n del factor rlm1. Adem\u00e1s se ha detectado uni\u00f3n in vivo de rlm1 al promotor del  gen fks1 y que dicha capacidad de uni\u00f3n no est\u00e1 regulada por la ruta pkc. por otro lado, se ha detectado que la map  quinasa sit2 de la ruta pkc se une al dna en la zona cercana al atg del gen fks1. Se detecto que la rna polimerasa  ii se une tanto a la zona del atg como al final del gen fks1 con independencia de la funci\u00f3n de la ruta pkc. Por otro  lado, se ha observado una modificaci\u00f3n post-traduccional de la proteina paf1 (miembro del complejo paf1 c)  dependiente de sit2, consistente con una posible fosforilacion de paf1 por sit2. Se observo que aunque la ruta no regula  la capacidad de union in viva al gen fks1 de paf1 en la zona del atg si parece controlar el desplazamiento de paf1 a 10  largo del gen fks1. En esta tesis se ha realizado un estudio de expresi\u00f3n global de un mutante pkc1-8 termosensible,  dicho estudio ha permitido identificar 65 genes cuya expresi\u00f3n se modifica significativamente al inactivar la funcion pkc1.  en este mismo sentido se han identificado prote\u00ednas asociadas a pkc1 y sit2 mediante la utilizaci\u00f3n del m\u00e9todo tap de  purificaci\u00f3n de prote\u00ednas. A destacar las prote\u00ednas rpc34, ssk22 y apd 1 que se detectaron asociadas a pkc1. Y las  prote\u00ednas mot1, sec31 y kap123 asociadas a sit2.
