Las enzimas implicadas en la din\u00e1mica degradativa del beta-glucano, componente mayoritario de la pared celular, son las beta-glucanasas, que catalizan la hidr\u00f3lisis del enlace beta-o-glicos\u00eddico de la cadena del carbohidrato. la primera parte de la tesis se centr\u00f3 en el estudio estructural de las (1,3)beta-endoglucanasas de la familia ghf81 de la levadura schizosaccharomyces pombe, speng1 y speng2, y saccharomyces cerevisiae sceng2. Se procedi\u00f3 a purificar estas prote\u00ednas y analizar diferentes variables bioqu\u00edmicas. Mediante hplc-ped se estudi\u00f3 su mecanismo hidrol\u00edtico demostrando su capacidad para degradar como m\u00ednimo un laminari-pentasac\u00e1rido, de manera similar a otros miembros de la familia ghf81. Se identific\u00f3 el posible dominio catal\u00edtico de sceng2 en su extremo carboxilo, el cual fue purificado posteriormente y analizado. Mediante mutag\u00e9nesis dirigida se identificaron varios amino\u00e1cidos clave en su actividad enzim\u00e1tica. Adem\u00e1s, se ha comprobado que speng1 posee un dominio de uni\u00f3n a glucanos en su extremo carboxilo; \u00e9ste fue purificado y se comprob\u00f3 su especificidad frente a sustratos insolubles con enlace (1,3)beta.  posteriormente, se estudi\u00f3 la implicaci\u00f3n de eng1 y agn1 ((1,3)alfa-endoglucanasa) en el proceso de separaci\u00f3n celular y los mecanismos de transporte y actuaci\u00f3n en las zonas donde son requeridas. Comprobamos que eng1 es necesaria para  degradar el septo primario que separa las dos c\u00e9lulas hijas tras la mitosis, y agn1 el cilindro de pared que rodea a este septo de separaci\u00f3n. Ambas prote\u00ednas se localizan en forma de anillo rodeando al septo en c\u00e9lulas silvestres. El estudio de su localizaci\u00f3n en mutantes defectivos en separaci\u00f3n celular, como los del complejo del exocisto, mid2  y spn , permiti\u00f3 concluir que en s.
