El cargado de la helicasa en el adn es un paso fundamental en el inico de la replicaci\u00f3n. Las helicasas son reclutadas en el adn gracias a prote\u00ednas primosomales que difieren en cada organismo.     en esta tesis se muestran las caracter\u00edsticas estructurales de diferentes complejos macromoleculares que tienen lugar en el inicio de la replicaci\u00f3n de la bacteria gram-positiva bacillus subtilis.     en primer lugar, se ha obtenido la estructura del complejo helicasa-cargador dnac-dnal mediante crio-microscopia electr\u00f3nica. Este complejo se organiza como una estructura toroidal donde tres d\u00edmeros de dnal se sit\u00faan sobre un anillo hexam\u00e9rico formado por la prote\u00edna dnac.     en segundo lugar se ha estudiado la estructura de dnab, el segundo cargador de la helicasa dnac, mediante diversas t\u00e9cnicas de elucidaci\u00f3n estructural. Dnab se organiza como un tetramero asim\u00e9trico cuya arquitectura recuerda al a del complejo gamma de escherichia coli. Esta semejanza, junto con la estructura del complejo dnac-dnal, sugiere que el cargado de la helicasa dnac se realiza mediante el mecanismo de apertura del anillo. Por \u00faltimo, se ha caracterizado tridimensionalmente el polimorfismo cuaternario de la helicasa. G40p ha permitido la localizaci\u00f3n de los diferentes dominios de esta familia de helicasas as\u00ed como determinar la implicaci\u00f3n de estos en el polimorfismo cuaternario. La transici\u00f3n entre los diferentes estados de simetr\u00eda ha sido propuesto como mecanismos para la translocaci\u00f3n de la helicasa sobre el adn.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos
