Entre ellas, la enfermedad de alzheimer es una de las m\u00e1s representativas debido a su importancia social y cl\u00ednica. Uno de los retos en la actualidad es el desarrollo de nuevos f\u00e1rmacos que frenen la evoluci\u00f3n de la enfermedad, o al menos palien sus s\u00edntomas. en esta tesis doctoral se ha estudiado la actividad protectora frente a diferentes condiciones t\u00f3xicas, de compuestos obtenidos mediante qu\u00edmica combinatoria. hemos puesto a punto un cultivo primario de neuronas de hipocampo de cerebro de rata, que nos permite realizar estudios hasta 3-4 semanas in vitro. Se ha analizado al evoluci\u00f3n de la excitotoxidad mediada por glutamato y nmda a lo largo de los div, lo que nos permite concluir que al 14 div la muerte provocada por glutamato est\u00e1 mediada principalmente a trav\u00e9s de los receptores nmda, y a 21 div aumenta la muerte neuronal que se produce v\u00eda otros receptores no-nmda. La muerte inducida por nmda a 14 div en neuronas de hipocampo es dependiente de ca2+, produce una morfolog\u00eda necr\u00f3tica en las c\u00e9lulas y cursa con la generaci\u00f3n de especies reactivas del ox\u00edgeno. Nmda induce la activaci\u00f3n de  la v\u00eda de supervivencia neuronal de creb, de forma dependiente del tiempo y de la concentraci\u00f3n de este agonista. Tambi\u00e9n hemos caracterizado la toxicidad del p\u00e9ptido beta-amiloide en neuronas de hipocampo. hemos ensayado varias n-alquilglicinas, obtenidas a partir de qu\u00edmica combinatoria, frente a excitotoxicidad mediada por nmda y glutamato en neuronas de hipocampo de cerebro de rata. varias de ellas, destacando n20c, son bloqueadores selectivos del receptor de nmda, con una moderada afinidad, y a myor efecto protector en choques exitot\u00f3xicos de varias horas. N20c bloquea el canal del receptor nmda, y protege de la generaci\u00f3n de especies reactivas del ox\u00edgeno y delos cambios en la fosfor<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis
