Recientemente se han caracterizado varios factores de la maquinaria de replicaci\u00f3n del mtdna, aunque el mecanismo b\u00e1sico de s\u00edntesis de las dos cadenas de dna est\u00e1 en discusi\u00f3n y se desconocen los elementos que regulan el proceso. El uso de d. Melanogaster como sistema modelo animal est\u00e1 constituyendo una herramienta experimental de gran utilidad para el estudio de los componentes que configuran la maquinaria basal de replicaci\u00f3n, mantenimiento y transcripci\u00f3n del mtdna. en nuestro grupo hemos analizado c\u00f3mo se regula la expresi\u00f3n de diversos genes que codifican factores esenciales para el metabolismo del mtdna en drosophila. Al contrario de lo que sucede con la subunidad catal\u00edtica de la dna polimerasa mitocondrial (pol    ), la expresi\u00f3n de la subunidad accesoria (pol    ) est\u00e1 sometida a una estricta regulaci\u00f3n transcripcional, y los niveles del mensajero pol     aumentan previamente a la activaci\u00f3n de la s\u00edntesis del mtdna durante el desarrollo embrionario. En esta tesis doctoral hemos profundizado en los mecanismos que controlan la expresi\u00f3n de este gen potencialmente clave en la regulaci\u00f3n de la replicaci\u00f3n mitocondrial.  hemos demostrado que pol  b     se expresa en drosophila en un mensajero bicistr\u00f3nico funcional que posee caracter\u00edsticas procari\u00f3ticas. Este mrna tambi\u00e9n codifica la subunidad c de la glutamil trnagln amidotransferasa (gatc). Hemos demostrado que gatc se localiza en la mitocondria y es esencial para el proceso de bios\u00edntesis de prote\u00ednas, sugiriendo que las mitocondrias animales utilizan una ruta de transamidaci\u00f3n para la s\u00edntesis del trnagln. diversas evidencias experimentales indican que gatc es una prote\u00edna bifuncional que tambi\u00e9n est\u00e1 implicada en la regulaci\u00f3n
