Esta patolog\u00eda causa importantes p\u00e9rdidas econ\u00f3micas en las piscifactor\u00edas, y no se dispone de un sistema de profilaxis eficaz. A pesar de la gran importancia de esta enfermedad, se conoce muy poco sobre los mecanismos de patogenicidad de f. Psychrophilum. En el presente trabajo se ha dise\u00f1ado un nuevo medio de cultivo s\u00f3lido para mejorar la recuperaci\u00f3n de c\u00e9lulas viables a partir del cultivo l\u00edquido, y se han desarrollado una serie de m\u00e9todos como la conjugaci\u00f3n, transformaci\u00f3n y vectores de expresi\u00f3n, que permiten la manipulaci\u00f3n gen\u00e9tica de f. psychrophilum. aplicando la tecnolog\u00eda desarrollada se han obtenido una serie de mutantes en caracteres descritos como implicados en la virulencia de f. Psychrophilum (motilidad, actividad proteol\u00edtica extracelular y limitaci\u00f3n de crecimiento en ausencia de hierro). De entre estos mutantes, en la presente memoria se estudi\u00f3 el denominado fp523, que present\u00f3 simult\u00e1neamente defectos en los tres rasgos fenot\u00edpicos anteriormente se\u00f1alados y una disminuci\u00f3n en su virulencia. Adem\u00e1s, este mutante mostr\u00f3 tambi\u00e9n alteraciones en el crecimiento y un aumento a la sensibilidad al dtt y, al contrario que la cepa parental, fue capaz de formar biofilm. El gen interrumpido en este mutante, denominado tlpb, codificaba una tioldisulfuro oxidorreductasa implicada en la reducci\u00f3n de puentes disulfuro en las prote\u00ednas
