Las dos caracteristicas fundamentales que definen la enfermedad de alzheimer (ea) son la formaci\u00f3n de unos acumulos extracelulares denominados placas seniles y de unos acumulos intracelulares que se denominan ovillos neurofibrilares. El p\u00e9ptido beta amiloide es el componente fundamental de las placas seniles y es un derivado de la prote\u00edna precursora del p\u00e9ptido beta amiloide o app. Desde que en 1991 se gener\u00f3 el primer transg\u00e9nico de esta prote\u00edna, se han realizado una gran variedad de animales transg\u00e9nicos que son capaces de reproducir parcialmente algunos de los sintomas, aunque la mayor\u00eda de ellos ha sido dise\u00f1ado para el est\u00fadio de la ea familiar, una variante minoritaria de la enfermedad. Nosotros hemos generado un nuevo modelo para el estudio de la enfermedad usando un cromosoma artificial de levaduras (yac), con un tama\u00f1o de 520 kb, que ha sido modificado mediane recombinaci\u00f3n hom\u00f3loga en levaduras para mantener el gen de app completo, con todas sus regiones reguladoras pero eliminando la presencia de otros genes, como el factor de transcripci\u00f3n gabpa, presentes en el yac de partida. usando el m\u00e9todo de transgenesis mediada por inyecci\u00f3n intracitoplasm\u00e1tica de cabezas de espermatozoides hemos sido capaces de generar varias l\u00edneas de ratones transg\u00e9nicos. En dos de estas l\u00edneas, la 35 y 64, se ha integrado el yac completo. El an\u00e1lisis de la expresi\u00f3n del gen y la prote\u00edna, ha resultada enormemente similar al del gen end\u00f3geno de rat\u00f3n.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis
