En concreto, las pgs renales contrarrestan la exacerbada actividad vasoconstrictora asociada a la activaci\u00f3n de los sistemas vasoactivos end\u00f3genos que tiene lugar en esta patolog\u00eda. De hecho, se ha podido observar que en pacientes cirr\u00f3ticos, la inhibici\u00f3n aguda de las pgs con aines se asocia un empeoramiento significativo en la funci\u00f3n renal, por lo que en la pr\u00e1ctica l\u00ednica los pacientes con cirrosis y ascitis no pueden recibir tratamientos prolongados con aines, debido al elevado riesgo de desarrollar fallo renal y ascitis regractaria.     a pesar de que el papel de las pgs en el mantenimiento de la funci\u00f3n renal en la cirrosis est\u00e1 bien establecido, se desconoce cu\u00e1l es la isoforma de la ciclooxigenasa (cox) responsable de la s\u00edntesis de pgs involucradas en la homeostasis renal en esta enfermedad. Con el prop\u00f3sito de esclarecer este aspecto, adem\u00e1s de caracterizar el efecto delos nuevos inhibidores selectivos de la cox-2, y especialmente si su uso puede o no causar efectos del et\u00e9reos renales en la cirrosis con ascitis, se ha llevado a cabo el presente proyecto. Los objetivos del mismo fueron:     1,- caracterizar la expresi\u00f3n y distribuci\u00f3n renal de cox-1 y cox-2 en ratas control y cirr\u00f3ticas.     2,- comparar los efectos producidos por la administraci\u00f3n de un inhibidor selectivo cox-1 (sc-560), con los de un inhibidor selectivo cox-2 (celexoxib), sobre la funci\u00f3n renal, respuesta diur\u00e9tica y natirur\u00e9tica a la furosemida y metabolismo del agua, en ratas con cirrosis y ascitis.     3,- investigar, in vitro, los mecanismos de acci\u00f3n de celecoxib en c\u00e9lulas renales, evaluando los efectos de celecoxib sobre la bios\u00edntesis de eicosanoides vasoactivos, la liberaci\u00f3n de renina, la activaci\u00f3n de ppargamma y la expresi\u00f3n de a-sma en c\u00e9lulas mesangiales de rata.     4,-
