La oms estima que el 3% de la poblaci\u00f3n mundial est\u00e1 cl\u00ednicamente afectada por este virus. M\u00e1s de la mitad de los casos progresan a infecciones cr\u00f3nicas que frecuentemente desarrollan cirrosis hep\u00e1tica que puede evolucionar a carcinoma hepatocelular. Sin embargo, el mecanismo de patog\u00e9nesis es a\u00fan poco conocido debido a la falta de un sistema de cultivo celular y un modelo viral adecuado para la infecci\u00f3n y replicaci\u00f3n del virus. El virus vaccinia (vv) ha sido un vector ampliamente usado para la expresi\u00f3n de prote\u00ednas heter\u00f3logas, por ello hemos desarrollado un nuevo sistema de expresi\u00f3n basado en un virus recombinante del virus vaccinia y que denominamos vt7-hcv7.9. Este virus recombinante expresa de forma inducible las prote\u00ednas estructurales (core, e1 y e2) y no estructurales (ns2, ns3, ns4a, ns4b, ns5a y parte de ns5b) del orf del subtipo 1b del hcv. An\u00e1lisis por microscop\u00eda confocal y por western blot han revelado que la poliprote\u00edna viral es eficientemente procesada en diferentes l\u00edneas celulares (hela, bsc40 y hepg2) infectadas con el virus recombinante vt7-hcv7.9, expres\u00e1ndose las diferentes prote\u00ednas estructurales y no estructurales de hcv. Adem\u00e1s, hemos visto que las prote\u00ednas de hcv expresadas a partir de vt7-hcv7.9 se localizan exclusivamente en el citoplasma con una distribuci\u00f3n puntuada, colocalizan con el ret\u00edculo endoplasm\u00e1tico (re) y las mitocondrias y produce rotura del aparato de golgi. En estudios por miscroscop\u00eda electr\u00f3nica de c\u00e9lulas humanas infectadas con el virus recombinante, se observ\u00f3 que las prote\u00ednas de hcv inducen alteraciones en la arquitectura de las c\u00e9lulas infectadas caracterizadas por la p\u00e9rdida de la organizaci\u00f3n del re, hinchamiento de las mitocondrias, formaci\u00f3n de red de membranas y de estructuras electr\u00f3n densas en la red de membrana, dispersas en varias \u00e1reas.
