{"id":79947,"date":"2018-03-09T23:25:50","date_gmt":"2018-03-09T23:25:50","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/regulacion-de-la-expresion-de-sdf1-alfa-cxcl12-efecto-de-ebna-1-tat-y-tax\/"},"modified":"2018-03-09T23:25:50","modified_gmt":"2018-03-09T23:25:50","slug":"regulacion-de-la-expresion-de-sdf1-alfa-cxcl12-efecto-de-ebna-1-tat-y-tax","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/ciencias-medicas\/regulacion-de-la-expresion-de-sdf1-alfa-cxcl12-efecto-de-ebna-1-tat-y-tax\/","title":{"rendered":"Regulaci\u00f3n de la expresi\u00f3n de sdf1 alfa \/ cxcl12. efecto de ebna 1, tat y tax."},"content":{"rendered":"<h2>Tesis doctoral de <strong>  Alonso Lobo Juan  Manuel <\/strong><\/h2>\n<p>El objetivo de este trabajo en un pricipio fue analizar la regulaci\u00f3n del promotor del cxcl12 en c\u00e9lulas productoras constitutivas: lc5, ms5, y u373, y determinar tanto los est\u00edmulos de inducci\u00f3n, como las regiones reguladoras implicadas en la regulaci\u00f3n de dicho promotor.  en una segunda parte, el objetivo fue analizar el efecto de la prote\u00edna tax de htlv-1 sobre la expresi\u00f3n de cxcl12 en linfocitos, as\u00ed como en c\u00e9lulas t de leucemias\/linfomas asociados a una infecci\u00f3n por htlv-1.  se observ\u00f3 que una expresi\u00f3n transitoria de la prote\u00edna tax en c\u00e9lulas jurkatt produjo una importante transactivaci\u00f3n del promotor de cxcl12. de acuerdo con estos resultados, se observ\u00f3 la presencia tanto de mrna como de prote\u00edna en c\u00e9lulas mt2 (l\u00ednea celular infectada de forma cr\u00f3nica por el htlv-1); todo ello visto por rt-pcr, citometr\u00eda de flujo y marcaje por inmunofluorescencia. Cxcl12 fue detectado tambi\u00e9n en el sobrenadante de c\u00e9lulas mt2 por t\u00e9cnicas de elisa y chequeada su actividad funcional por ensayos de quimiotaxis.  biopsias de pacientes infectados con htlv-1 mostraron linfocitos cd3+ positivos para la expresi\u00f3n de cxcl12, mientras que no aparec\u00eda esta expresi\u00f3n en ganglios linf\u00e1ticos de pacientes no infectados por el htlv-1.  en base a estos resultados se puede concluir que la prote\u00edna tax de htlv-1 induce la expresi\u00f3n de la quimiocina cxcl12 a nivel transcripcional, apareciendo as\u00ed una expresi\u00f3n aberrante de la prote\u00edna cxcl12 tanto en l\u00edneas celulares infectadas con el htlv-1 como en linfocitos de biopsias provenientes de linfomas asociados a  este virus. El incremento en la expresi\u00f3n de cxcl12 por la infecci\u00f3n htlv-1 puede contribuir al reclutamiento de linfocitos sanos hacia los ganglios infectados por el htlv-1, induciendo as\u00ed la
