Se ha evidenciado la importancia de la fuerza i\u00f3nica del enzimoinmuno ensayo sobre su eficacia diagn\u00f3stica, siendo mucho m\u00e1s espec\u00edfica y sensible para el diagn\u00f3sitco de lupus eritematosos sist\u00e9mico la cuantificaci\u00f3n de anticuerpos de elevada avidez en condiciones de elevada fuerza i\u00f3nica. Se ha puesto de manifiesto la utilidad restringida del standard internacional anti-dsdna wo-80 de la oms solo como calibrador para ensayos realizados en condiciones de elevada fuerza i\u00f3nica y para la cuantificaci\u00f3n exclusiva de anticuerpos anti-dsdna de elevada avidez. El elosa desarrollado presenta, respecto a la inmunofluorescencia indirecta sobre crithidia lucilae, valores de sensibilidad y especificidad diagn\u00f3stica del 95,4 y 87% respectivamente. Considerando un valor discriminante de 45 ui\/ml.  se ha desarrollado un elisa cualitativo para la detecci\u00f3n de anticuerpos dirigidos contra las prote\u00ednas sm, u1-rnp, scl-70, juo-1, ss-a y ss-b del extracto nuclear soluble (ena), utilizando como substrato antig\u00e9nico una mezcla de p\u00e9ptidos nativos y recombinantes. Las condiciones de sensibilizaci\u00f3n de las microplacas se han optimizado mediante estrategia multivariante, con un dise\u00f1o experimental factorial de composici\u00f3n central y utilizando la metodolog\u00eda de las superficies de respuesta para la toma de decisiones sobre las condiciones \u00f3ptimas de sensibilizaci\u00f3n.
