Las neurotoxinas clostridiales tienen una elevada especifidad por diversas porte\u00ednas de membrana responsables de la fusi\u00f3n de membranas, impidiendo de esta manera la liberaci\u00f3n de neurotransmisores contenidos en las ves\u00edculas sin\u00e1pticas. Se asume que esta actividad es la base de su acci\u00f3n t\u00f3xica, aunque se han descrito otras actividades de la tetx, como la inducci\u00f3n de la activaci\u00f3n y posterior prote\u00f3lisis (down-regulation) de la porte\u00edna quinasa c (pkc) de la que existen diferentes isoformas. el objetivo de esta tesis ha sido la determinaci\u00f3n del efecto de la tetx sobre diferentes isoformas okc, su acci\u00f3n sobre v\u00edas de transducci\u00f3n de se\u00f1ales en que esta participa y la comparaci\u00f3n con el efecto del tpa y del ngf (factor de crecimiento nervioso).  el tratamiento de cultivos primarios de neuronas corticales fetales de rata con tpa (12-orto-tetradecanoil forbol-13-acetato) o con tetx induce la translocaci\u00f3n de la pkc a la fracci\u00f3n de membrana, fen\u00f3meno que se asocia con su activaci\u00f3n. Utilizando anticuerpos  espec\u00edficos contra seis isoformas pkc (&#8212;&#8211;) se demuestra que el tpa induce la translocaci\u00f3n de todas las isoformas excepto las e y \u00ed\u00a7. La tetx induce translocaci\u00f3n de la pkcb, mientras que induce down-regulation de las isoformas a, y,- y e. No presenta ning\u00fan efecto sobre la pkc\u00ed\u00a7. Esta activaci\u00f3n de la pkcb se correlaciona con la estimulaci\u00f3n de fosfolipasas de tipo c por la tetx.  en una segunda parte se han utilizado sinaptosomas como sistema de trabajo. el tratamiento de estos con tpa induce down-regulation de las isoformas a, b y  y, as\u00ed como de la pkce, mientras que la pkc8 experimenta translocaci\u00f3n sostenida a altas concentraciones de tpa.
