{"id":81486,"date":"1999-03-12T00:00:00","date_gmt":"1999-03-12T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/estudio-de-la-proliferacion-de-las-celulas-de-fibrosarcoma-que-sobreexpresan-la-mangeso-superoxido-dismutasa\/"},"modified":"1999-03-12T00:00:00","modified_gmt":"1999-03-12T00:00:00","slug":"estudio-de-la-proliferacion-de-las-celulas-de-fibrosarcoma-que-sobreexpresan-la-mangeso-superoxido-dismutasa","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/estudio-de-la-proliferacion-de-las-celulas-de-fibrosarcoma-que-sobreexpresan-la-mangeso-superoxido-dismutasa\/","title":{"rendered":"Estudio de la proliferacion de las celulas de fibrosarcoma que sobreexpresan la mangeso superoxido dismutasa."},"content":{"rendered":"<h2>Tesis doctoral de <strong>  Rodriguez De Ledesma Vega Ana M. <\/strong><\/h2>\n<p>Los estudios realizados en nuestro laboratorio demuestran que la inhibici\u00f3n proliferativa asociada a la sobreexpresi\u00f3n de la manganeso super\u00f3xido dismutasa (mnsod) queda parcialmente revertida en una atm\u00f3sfera baja de ox\u00edgeno (3%). a pesar de que numerosos estudios demuestran que la sobrexpresi\u00f3n de la mnsod se asocia a una p\u00e9rdida del fenotipo maligno in vitro y disminuye la capacidad de formar tumores in vivo, no est\u00e1 claro c\u00f3mo la mnsod afecta a la proliferaci\u00f3n celular.  dentro de la mitocondria, la mnsod cataliza la reacci\u00f3n de simutaci\u00f3n del super\u00f3xido a per\u00f3xido de hidr\u00f3geno (h2o2), el cual es posteriormente metabolizado por acci\u00f3n de las enzimas catalasa o dela glutati\u00f3n peroxidasa. En el presente trabajo hemos estudiado si la inhibici\u00f3n proliferativa asociada a la sobreexpresi\u00f3n de la mnsod es debido a un aumento del producto de dismutaci\u00f3n, el h2o2. para ellos hemos sobreexpresado la catalasa en c\u00e9lulas que sobreexpresan altos niveles de mnsod (ht15) con el uso de pl\u00e1smidos recombinantes para dirigir la catalasa a la \u00f1mitocondria y al citosol. La sobreexpresi\u00f3n de la catalasa tanto en la mitocondria como es el citosol reverti\u00f3 la inhibici\u00f3n proligerativa y clonog\u00e9nica asociada a la sobrexxpresi\u00f3n de la mnsod. Sin embargo, el uso de sondas reductoras no nos ha permitido demostrar un aumento de per\u00f3xidos intracelulares en las c\u00e9lulas que sobreexpresan la catalas. nuestros resultados tambi\u00e9n demustran que le consumo de ox\u00edgeno aumenta en todas las c\u00e9lulas que sobreexpresan la catalasa independientemente de que la actividad de la mnsod est\u00e9 aumentada. Los niveles de atp, que est\u00e1n significativamente disminuidos en las c\u00e9lulas que sobreexpresan la mnsod, son adem\u00e1s, revertidos a niveles normales con la sobreexpresi\u00f3n de la catalasa. por otro lado, la sobreexpresi\u00f3n de la mnsod aumenta la sensibilidad de las c\u00e9lulas a los dadores del \u00f3xido n\u00edtrico (no\u00c2\u00ba).
