{"id":81561,"date":"1999-10-12T00:00:00","date_gmt":"1999-10-12T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/asimilacion-de-nitrato-en-la-leguminosa-modelo-lotus-japonicus\/"},"modified":"1999-10-12T00:00:00","modified_gmt":"1999-10-12T00:00:00","slug":"asimilacion-de-nitrato-en-la-leguminosa-modelo-lotus-japonicus","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/asimilacion-de-nitrato-en-la-leguminosa-modelo-lotus-japonicus\/","title":{"rendered":"Asimilaci\u00f3n de nitrato en la leguminosa modelo lotus japonicus."},"content":{"rendered":"<h2>Tesis doctoral de <strong> Alicia Orea Garcia <\/strong><\/h2>\n<p>Lotus japonicus es una leguminosa que gracias a sus caracter\u00edsticas bioqu\u00edmicas, gen\u00e9ticas y fisiol\u00f3gicas ha sido considerada como planta modelo. se ha estudiado la fisiolog\u00eda de la asimilaci\u00f3n de nitrato en esta planta mediante el uso de plantas transg\u00e9nicas, modificadas en la expresi\u00f3n de la nitrato reductasa(nadh-nr, primera enzima que participa en la asimilaci\u00f3n de nitrato), as\u00ed como la clonaci\u00f3n y estudios de expresi\u00f3n de la nitrito reductasa (fd-nir,segunda enzima que participa en el proceso de la asimilaci\u00f3n de nitrato), y se ha desarrollado un programa de mutag\u00e9nesis convencional para la b\u00fasqueda de mutantes afectados en la asimilaci\u00f3n de nitrato. La asimilaci\u00f3n de nitrato en lotus japonicus tiene lugar principalmente en las raices pero, a diferencia de lo descrito para leguminosas de clima templado, grupo al cual pertenece, el aumento de la concentraci\u00f3n de nitrato en el medio de cultivo no conduce a un desplazamiento de la asimilaci\u00f3n de nitrato hacia la parte aerea de la planta. Mediante manipulaci\u00f3n con plantas transgenicas se ha conseguido el cambio de expresi\u00f3n de la nitrato reductasa en l. Japonicus, pero en ning\u00fan caso la actividad nitrato reductasa de hoja superaba a la de ra\u00edces. Se postula, pues, la existencia de un mecanismo de regulaci\u00f3n en esta planta que la imposibilita para asimilar una mayor cantidad de nitrato en hojas. Por otra parte, la clonaci\u00f3n de la nitrito reductada de raices de l.Japonicus ha revelado que no es necesario una estructura o secuencia particular en las fd-nir de raices diferente a las de hojas, para poder funcionar con el donador de electrones de raices, hecho que se ve apoyado por la existencia de un unico gen por genoma haploide en l. Japonicus. Sin embargo si se observan diferencias en cuanto a la regulaci\u00f3n de la expresi\u00f3n por luz de esta enzima, respecto a las de plantas asimiladoras en hojas.
