Se ha sugerido que la pineal de los mamiferos tiene influencia sobre la funci\u00f3n del sistema nervioso central y sobre la actividad de la nos de tipo constitutivo. La melatonina interacciona con la calmodulina, y esta uni\u00f3n produce una inhibici\u00f3n de la actividad nos en el hipot\u00e1lamo de rata de una manera dependiente de la dosis.  los estudios in vitro muestran que la concentraci\u00f3n fisiologica de melatonina(1nm)inhibe la producci\u00f3n de pge2 y ampc\u00edclico inducida por na en hipot\u00e1lamo medio-basal (mbh) de la rata.  por otra parte, la noradrenalina induce la liberaci\u00f3n de lhrh estimulando la produccion de \u00f3xido nitrico. El no activa la ciclooxigenasa, que sintetiza pge2,ampc y la liberaci\u00f3n de lhrh parece estar mediado por la inhibici\u00f3n de la actividad \u00f3xido n\u00edtrico sintasa. Estos resultados sugieren la existencia de un nuevo mecanismo por el cual la melatonina puede regular la fisiolog\u00eda de la unidad hipot\u00e1lamo-pituitaria.  los resultados demuestran tambi\u00e9n que el cgp 52608 que es un an\u00e1logo de la melatonina al receptor nuclear es capaz de reproducir los efectos de la melatonina sobre la producci\u00f3n de pge2 y la liberacion de lhrh en la parte medio basal del hipot\u00e1lamo de rata. Sin embargo, cuando se incub\u00f3 el homogenado del hipot\u00e1lamo con cgp no se observ\u00f3 un efecto inhibidor de la actividad nos. Estos resultados identifican la existencia de receptores nucleares de melatonina en el hipot\u00e1lamo.  la adici\u00f3n del an\u00e1logo de melatonina para el receptor de la membrana s 20098 a concentraci\u00f3n fisiol\u00f3gica en el medio de cultivo de mbh de rata produjo una inhibici\u00f3n de la liberaci\u00f3n de lhrh, pero no se observ\u00f3 ning\u00fan efecto inhibidor en la
