Pta es una peque\u00f1a prote\u00edna nuclear de aproximadamente 12kd., Sumamente ac\u00eddica cuya funci\u00f3n es desconocida. en diversos modelos experimentales se ha visto que existe una correlaci\u00f3n entre los niveles de pta y su actividad proliferativa, as\u00ed los niveles de pta son mayores en c\u00e9lulas que se dividen m\u00e1s activamente como tambi\u00e9n en tumores, y por ello, es utilizado como marcador tumoral.  en el presente trabajo caracterizamos inmunol\u00f3gicamente el anticuerpo anti-pta a fin de desarrollar t\u00e9cnicas como el western blot y la inmunofluorescencia. Una vez caracterizado, y tras poner a punto t\u00e9cnicas como la inmunofluorescencia, encontramos que la expresi\u00f3n de pta disminuye en c\u00e9lulas en quiescencia. Mediante microscop\u00eda electr\u00f3nica observamos como en mitosis (profase tard\u00eda) la prote\u00edna disminuye en el citoplasma. La posibilidad de que esta disminuci\u00f3n se debiera a la proteolisis de la prote\u00edna, nos indujo a estudiar la degradaci\u00f3n de pta. observamos que la actividad proteol\u00edtica depende del estad\u00edo celular, siendo mayor en mitosis y en c\u00e9lulas serodeprivadas. Esta degradaci\u00f3n de pta en las c\u00e9lulas en mitosis es debida a una ciste\u00edn proteasa neutra, no liosomal, dependiente de calcio (una calpa\u00edna). Ambas isoformas micro y milicalpa\u00edna degradan pta. La degradaci\u00f3n de pta por calpa\u00edna \u00abin vitro\u00bb origina tres fragmentos, localiz\u00e1ndose los puntos de corte en la regi\u00f3n n-terminal como se observ\u00f3 mediante western blot y espectrofotometr\u00eda de masas (maldi-tof). Uno de los puntos de corte podr\u00eda ser el responsable de la obtenci\u00f3n de la forma des(25-28) timosina alfa1. Este sistema de degradaci\u00f3n podr\u00eda ser el responsable de la degradaci\u00f3n \u00abin vivo\u00bb dada que el mismo patr\u00f3n de degradaci\u00f3n fue obtenido en extratos celulares.
