Se ha demostrado que estos compuestos son esenciales para el crecimiento y desarrollo del par\u00e1sito protozoo trypanosoma cruzi, agente causal de la enfermedad de chagas. Para llegar a un mayor conocimiento de la regulaci\u00f3n e importancia biol\u00f3gica de esta enzima, el trabajo realizado en esta tesis, ha consistido en:  a) el an\u00e1lisis y caracterizaci\u00f3n del gen en trypanosoma cruzi, demostr\u00e1ndose que el gen tiene una secuencia de 1305 pb, es de copia \u00fanica, se localiza en un cromosoma de unas 2.1mb y se transcribe dando lugar a un mrna de 2.2kb. La prote\u00edna para la que codifica este gen tiene una secuencia deducida de 435 amino\u00e1cidos, estando altamente conservada con respecto al dominio citos\u00f3lico de otras hmgrs eucariotas.  b) establecimiento de actividad hmgr en t. cruzi, comprob\u00e1ndose que est\u00e1 presente en estos par\u00e1sitos y que se asocia a una fracci\u00f3n soluble obtenida por centrifugaci\u00f3n diferencial.  c) el desarrollo de un sistema de expresi\u00f3n heter\u00f3logo de la enzima de t. Cruzi en c\u00e9lulas de e.Coli y purificaci\u00f3n de la proteina recombinante.  d) estudios de reemplazamiento g\u00e9nico, que dieron como resultado la aparici\u00f3n de mutantes con nuevos elementos extracromos\u00f3micos que incorporan copias del gen sin lograrse mutantes que caracieran del gen salvaje.  e) el desarrollo de un sistema de expresi\u00f3n hom\u00f3logo, para poder llevar a cabo estudios de localizaci\u00f3n intracelular de la enzima y de sensibilidad del par\u00e1sito a inhibidores competitivos de la hmgr, estatinas. Los resultados mostraron la sobreexpresi\u00f3n de la enzima del orden de 4 veces, los par\u00e1sitos mutantes presentan resistencia aumentada a la lovastatina y simvastatina.  f) estudios de localizaci\u00f3n intracelular de la enzima hmgr de t.
