En este microorganismo, la ruta del alfa-aminoadipato juega un papel adicional, adem\u00e1s de la bios\u00edntesis de lisina, ya que suministra alfa-aminoadipato para la formaci\u00f3n de penicilina. El primer paso de la ruta de bios\u00edntesis de lisina, la formaci\u00f3n de homocitrato por la enzima homocitrato sintasa, parece ser un punto clave en la regulaci\u00f3n del flujo metab\u00f3lico a trav\u00e9s de la ruta. En esta memoria se recoge la caracterizaci\u00f3n gen\u00e9tica y molecular de este paso enzim\u00e1tico.  el gen que codifica la homocitrato sintasa, primera enzima de laruta de formaci\u00f3n de lisina, de p. Chrysogenum fue clonado, mediante hibridaci\u00f3n heter\u00f3loga, y posteriormente secuenciado. Mediante electroforesis en geles de pulsos se separaron los cromosomas de varias estirpes de p. Chrysogenum, y mediante hibridaci\u00f3n se determin\u00f3 la localizaci\u00f3n cromos\u00f3mica del gen lys1 en las cepas estudiadas. La funci\u00f3n del gen lus1, como gen que codifica la homocitrato sintasa, fue demostrada mediante la obtenci\u00f3n de tres mutantes interrumpidos en dicho gen, mediante la t\u00e9cnica de doble recombinaci\u00f3n. Los tres mutantes exhibieron auxotrof\u00eda de lisina y, \u00fanicamente, niveles residuales de actividad homocitrato sintasa.  se estudi\u00f3 mediante hibridaci\u00f3n de adn total de tres cepas de p. Chrysogenum, el n\u00famero de copias del gen lys1 en el genoma, comprob\u00e1ndose la existencia de una sola copia del gen lys1 en las tres cepas objeto de estudio. Esto, junto a los resultados obtenidos en los experimentos de interrupci\u00f3n gen\u00e9tica, demostr\u00f3 que solamente existe un gen que codifique la enzima homocitrato sintasa en p. Chrysogenum.  en saccharomyces cerevisiae existen dos isoenzimas de la homocitrato sintasa, ambas localizadas en el n\u00facleo celular.
