Objetivos: implantar en el hospital universitario san carlos las t\u00e9cnicas de estudio molecular de los reordenamientos y las deleciones de genes supresores de tumor m\u00e1s frecuentemente observados en las leucemias agudas. Describir la frecuencia y correlaci\u00f3n morfol\u00f3gica e inmunofenot\u00edpica de estas alteraciones moleculares. Describir la utilidad de la rt-pcr para detectar enfermedad m\u00ednima residual (emr) a trav\u00e9s del an\u00e1lisis de los reordenamientos bcr\/abl y pml\/raralfa. comparar la detecci\u00f3n de alteraciones gen\u00e9ticas mediante t\u00e9cnicas de biolog\u00eda molecular con los hallazgos obtenidos a trav\u00e9s de la citogen\u00e9tica convencional. Relacionar la presencia de estos marcadores moleculares con las caracter\u00edsticas cl\u00ednicas de los pacientes al diagn\u00f3stico y con su evoluci\u00f3n. material y m\u00e9todos: se han estudiado 113 pacientes con leucemia aguda. Se han estudiado los genes mll, p16 y p15 mediante southern blot. Se han analizado mediante rt-pcr los reordenamientos bcr\/abl, tel\/aml1, e2a\/pbx1, mll\/af4, mll\/af6, mll\/af9, mll\/enl, mll\/ell, duplicaci\u00f3n del gen mll, pml\/raralfa, aml1\/eto y cbfb\/myh11. Resultados y conclusiones: sobre un total de 113 pacientes con leucemia aguda, el an\u00e1lisis molecular se ha podido realizar en la inmensa mayor\u00eda de los casos, 110 (97.3%). El estudio de las alteraciones moleculares investigadas ha permitido detectar alg\u00fan reordenamiento y\/o deteci\u00f3n en casi la mitad de los pacientes, 43.6% del total y 47,4% de los casos en edad pedi\u00e1trica. es m\u00e1s de la mitad de los casos con alteraciones moleculares en los que el cariotipo fue valorable (57.9%)no se encontr\u00f3 la alteraci\u00f3n
