Paz <\/strong><\/h2>\n<p>Los receptores de nmda est\u00e1n constituidos por dos tipos de subunidades, nr1 y nr2. Se conocen cuatro isoformas de la subunidad nr2 (a-d). El receptor de nmda sufre un proceso de desensibilizaci\u00f3n en presencia prolongada de agonista, el cual es espec\u00edfico de la subunidad nr2 presente. As\u00ed los heter\u00f3meros nr1\/2a presentan marcada desensibilizaci\u00f3n en tanto que los nr1\/2c no. Estos receptores tambi\u00e9n presentan un proceso de desensibilizaci\u00f3n que es ocluido por glicina y la sensibilidad para este proceso depende igualmente de la subunidad nr2 presente. La subunidad nr1 contiene el sitio de uni\u00f3n a glicina, mientras que la subunidad nr2 alberga el sitio de uni\u00f3n al glutamato. Con el fin de comprender los mecanismos moleculares responsables de la desensibilizaci\u00f3n, tanto independiente como dependiente de glicina, hemos construido quimeras nr2a\/nr2c, que han sido coexpresadas transitoriamente con la subunidad nr1 salvaje en la l\u00ednea celular hek-293, y caracterizadas electrofisiol\u00f3gicamente mediante la t\u00e9cnica de \u00abpatch-clamp\u00bb y un sistema r\u00e1pido de perfusi\u00f3n.  en cuanto a la desensibilizaci\u00f3n glicina-independiente los resultados de las quimeras analizadas indicaron la existencia de dos procesos de desensibilizaci\u00f3n con diferentes cin\u00e9ticas y mediados por distintas partes de la mol\u00e9cula. Un mecanismo es r\u00e1pido y ligado a la regi\u00f3n n-terminal y el otro es m\u00e1s lento y viene determinado por residuos situados en la regi\u00f3n pre-m1 (4 amino\u00e1cidos justo antes del primer dominio trasnmembrana). La ausencia de desensibilizaci\u00f3n se observ\u00f3 en la subunidad nr2a que conten\u00eda ambos segmentos (n-terminal y pre-m1) de la subunidad nr2c.  la diferencia de afinidad por la glicina entre heter\u00f3meros nr1\/nr2a y nr1\/nr2c (posible base de la desensibilizaci\u00f3n glicina-dependiente) viene determinada por la regi\u00f3n n-terminal.
