Aunque sus propiedades farmacocin\u00e9ticas (fc) y farmacodin\u00e1micas (fd) han sido estudiadas en humanos y animales, no se sabe demasiado sobre la contribuci\u00f3n de su metabolito activo (norketamina) al efecto farmacol\u00f3gico. El objetivo de este estudio ha sido explorar la posible contribuci\u00f3n de la norketamina en el efecto farmacol\u00f3gico medido tras la administraci\u00f3n de ketamina a ratas. El estudio farmacocin\u00e9tico y farmacodin\u00e1mico se realiz\u00f3 en gurpos diferentes de ratas. En ambos estudios se administraron a ratas dosis de 65, 120 y 200 mg\/kg en infusiones de 25, 25 y 45 minutos, respectivamente. La determinaci\u00f3n simult\u00e1nea de las concentraciones de ketamina y norketamina en plasma se realiz\u00f3 a trav\u00e9s de cromatograf\u00eda luida de alta resoluci\u00f3n. La medici\u00f3n del efecto farmacol\u00f3gico se efectu\u00f3 a trav\u00e9s de la variaci\u00f3n en amplitud de onda del potencial evocado auditivo. El estudio demostr\u00f3 que la medici\u00f3n del potencial evocado auditivo es una forma fiable y objetiva para medir el efecto farmacol\u00f3gico de la ketamina. El modelo fc-fd seleccionado fue el modelo de efecto m\u00e1ximo sigmoide.
