En este trabajo, hemos observado que la eficiencia de transducci\u00f3n de tumores primarios y metast\u00e1sicos de h\u00edgado es muy baja cuando el vector se aplica intravascularmente. En el modelo de hcc multifocal inducido por dena, los hcc mayores de 2-5 mm son muy dif\u00edciles de transducir tanto si el adenovirus se administra por la vena porta como por la arteria hep\u00e1tica. La resistencia a ser transducidos parece estar relacionada con el desarrollo de una barrera entre la sangre y los hepatocitos tumorales. Nuestros resultados de microscop\u00eda electr\u00f3nica demuestran que esta barrera parece estar formada por la presencia de un endotelio grueso, carente de fenestraciones y con existencia de una capa de matriz extracelular entre el endotelio y las c\u00e9lulas tumorales. La administraci\u00f3n por la arteria hep\u00e1tica de compuestos vasoactivos, especialmente nitroglicerina, previa a la infusi\u00f3n del adenovirus, aumenta la eficiencia de transducci\u00f3n de hcc grandes.
