El molde cromatinico utilizado, el fragmento de dna nx1p7 de 239 pb, consta del promotor de la rna polimerasa t7 fusionado a la secuencia nx1 de origen nucleosomal. El analisis de los complejos histonas-dna nx1p7 en geles no desnaturalizantes revela un patron de reconstitucion altamente definido. El estudio de footprinting de dichos complejos confirma el posicionamiento del nucleosoma sobre nx1. La transcripcion del molde cromatinico en condiciones de fuerza ionica cercana a la fisiologica provoca la disociacion de las histonas del dna molde, la acumulacion consiguiente de dna libre y la desaparicion de las bandas de reconstituidos nucleosomales. Estos cambios en el patron de reconstitucion inicial son inherentes al proceso transcripcional ya que en ausencia de uno solo de los rntps o de la rna polimerasa los complejos nucleosomales permanecen inalterables. La disociacion de los complejos histonas-dna se acentua al transcribir en presencia de dna o cromatina competidores.  las histonas liberadas por efecto de la transcripcion se asocian al dna o a la cromatina presentes en el sistema como histonas en exceso permitiendo asi el avance de la rna polimerasa durante la transcripcion.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
