Josefa <\/strong><\/h2>\n<p>El objetivo de esta tesis ha consistido en el estudio de la din\u00e1mica del calcio en el ret\u00edculo endopl\u00e1smico de distintas c\u00e9lulas en respuesta a diversos est\u00edmulos. Para llevar a cabo las determinaciones experimentales utilizamos una t\u00e9cnica nueva basada en una fotoprote\u00edna, denominada aequorina, que emite luminiscencia cuando se une a este i\u00f3n de foma proporcional a su concentraci\u00f3n, dirigida espec\u00edficamente al interior del ret\u00edculo endopl\u00e1smico mediante ingenier\u00eda gen\u00e9tica y mutada para reducir su afinidad por calcio al rango de medida adecuado.  comenzamos nuestro estudio en la l\u00ednea celular tumoral hela, poniendo a punto la t\u00e9cnica de medida y determinando tanto el nivel estable como la din\u00e1mica de la (ca2+) en el ret\u00edculo. Adem\u00e1s evidenciamos el funcionamiento y los mecanismos de regulaci\u00f3n por calcio citos\u00f3lico, luminal y procesos redox del receptor del ip3.  posteriormente ampliamos el estudio de la homeostasis del calcio en ret\u00edculo endopl\u00e1smico a otros tipos celulares expresendo la aequorina mutada en el ret\u00edculo mediante vectores virales.  una vez comprobada la eficacia funcional de la t\u00e9cnica de infecci\u00f3n viral, centramos nuestro estudio enun tipo celular de gran relevancia fisiol\u00f3gica, por ser de estirpe neuroendocrina, la c\u00e9lula cromaf\u00edn.
