{"id":83398,"date":"2018-03-10T00:07:31","date_gmt":"2018-03-10T00:07:31","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/marcaje-de-afinidad-y-estudios-estructurales-de-centros-activos-de-glicosidasas-lactasa-intestinal-y-otras-glicosidasas-homologas\/"},"modified":"2018-03-10T00:07:31","modified_gmt":"2018-03-10T00:07:31","slug":"marcaje-de-afinidad-y-estudios-estructurales-de-centros-activos-de-glicosidasas-lactasa-intestinal-y-otras-glicosidasas-homologas","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/marcaje-de-afinidad-y-estudios-estructurales-de-centros-activos-de-glicosidasas-lactasa-intestinal-y-otras-glicosidasas-homologas\/","title":{"rendered":"Marcaje de afinidad y estudios estructurales de centros activos de glicosidasas: lactasa intestinal y otras glicosidasas homologas."},"content":{"rendered":"<h2>Tesis doctoral de <strong>  Diaz Arribas Juan  Carlos <\/strong><\/h2>\n<p>La lactasa intestinal (tambi\u00e9n denominada complejo lactasa-floriz\u00edn hidrolasa o lph) es una glicosidasa localizada en la membrana  de los enterocitos del intestino delgado de mam\u00edferos proyect\u00e1ndose hacia el l\u00famen del mismo. esta enzima posee dos centros activos diferentes localizados en dos regiones hom\u00f3logas de la misma cadena polipept\u00eddica: uno de ellos encargado de la hidr\u00f3lisis de la lactosa (el az\u00facar m\u00e1s de la leche) y el otro, aunque actualmente se desconoce su sustrato natural, viene siendo tradicionalmente ensayado con el glic\u00f3sido denominado florizina.  en esta tesis, mediante el empleo de inhibidores irreversibles de glicosidasas del tipo 2-desoxi-2-fluoroglic\u00f3sidos y el concurso de la espectroscop\u00eda de masas con ionizaci\u00f3n asistida por electrospray (esi-ms), ha sido posible asignar inequ\u00edvocamente cada una de las dos actividades catal\u00edticas de la lactasa intestinal de cordero lechal, lactasa y floriz\u00edn-hidrolasa, a cada uno de los dos centros activos de la enzima: la actividad lactasa el centro localizado en la regi\u00f3n iv de la enzima y la actividad floriz\u00edn-hidrolasa al centro presente en la regi\u00f3n iii.  as\u00edmismo, mediante experimentos de rmn de \u00abefecto nuclear overhauser transferido\u00bb (tr-noe), se ha realizado el estudio de la conformaci\u00f3n que en disoluci\u00f3n adopta un an\u00e1logo no hidrolizable de la glucosidasa blga de paenibacillus polymyxa dentro del centro activo de la misma. La informaci\u00f3n obtenida de estos estudios es \u00fatil para conocer el mecanismo catal\u00edtico de estas enzimas y para el dise\u00f1o de inhibidores y an\u00e1logos de sustrato con diferentes
