Esta prote\u00edna, de 27 kda, es ub\u00edcua y muy abundante en el n\u00facleo. est\u00e1 compuesta por tres dominios distintos: los dominios a(n-ter) y b(central) son similares y contienen el motivo hmg-box; el dominio c(c-ter), de car\u00e1cter ac\u00eddico, esta desestructurado. Se ha sugerido que la funci\u00f3n de hmg1 estar\u00eda relacionada con la regulacion de la transcripci\u00f3n dirigida por rna polimerasa ii. Se analiz\u00f3 la interacci\u00f3n de la hmg1 con los primeros intermediarios nucleoproteicos del complejo de preiniciaci\u00f3n(o pic) sobre un promotor basal. Se observ\u00f3 que la hmg1 interacciona directamente con los complejos tbp-promotor y tfiib-tbp-promotor en emsa. Hmg1 estabiliza la uni\u00f3n de tbp al dna e interfiere en la uni\u00f3n de tfiib en el complejo. Tfiia y hmg1 compiten entre si por interaccionar con tbp. Por esta raz\u00f3n, tfiia evita la interacci\u00f3n de hmg1 con los complejos tfiia-tbp-promotor y tfiia-tfiib-tbp-promotor. el conjunto de resultados muestran que hmg1 afecta la formaci\u00f3n del pic en un promotor basal: debido a la uni\u00f3n de hmg1 a tbp,tfiib no se une correctamente en el complejo, impidi\u00e9ndose por tanto la formacion correcta del pic. Este efecto podr\u00eda ser contrarrestado por tfiia.  la interacci\u00f3n hmg1-tbp parecer\u00eda ser especifica de especie, puesto que hmg1 interacciona con tbp humana(htbp), pero no con tbp a trav\u00e9s de la h\u00e9lice-a h2\u00c2\u00bfdel dominio conservado core. Los residuos ala319,asn327 y gly334 est\u00e1n directamente implicados en la interacci\u00f3n primaria con hmg1,siendo la contribuci\u00f3n del residuo asn327 la m\u00e1s importante. Estos residuos serian los responsables de determinar la especificidad de la interaci\u00f3n con hmg1. hmg-box a interacciona con htfiid en un extracto nuclear crudo de c\u00e9lulas hela, pero no con los complejos sl1 y tfiiib.
