A partir de la cepa virulenta b.Abortus 2308 se construyeron mediante transposici\u00f3n unos mutantes, que manteniendo un lipoposac\u00e1rido (lps) de tipo liso, presentaban las propiedades de la ec alteradas. Dichos mutantes fueron m\u00e1s sensibles a p\u00e9ptidos cati\u00f3nicos (polimixina b, poli-l-lisina y melitina), antibi\u00f3ticos y surfactantes (sds,deoxicolato y sarkosil), y exhibieron una mayor hidrofobicidad superficial. La capacidad de invasion en c\u00e9lulas hela y macrofagos result\u00f3 ser significativamente menor que la de la cepa parental, y las pocas bacterias capaces de penetrar en las c\u00e9lulas eucariotas eran r\u00e1pidamente destruidas. De igual modo, los estudios realizados en rat\u00f3n mostraron que los mutantes eran avirulentos. el hecho de que la velocidad de crecimiento fuera similar a la de la cepa parental demuestra que los genes alterados no son esenciales para la bacteria y que estan relacionados directamente con la virulencia. El an\u00e1lisis genetico mostr\u00f3 que el transpos\u00f3n se localizaba en los mutantes en dos genes adyacentes (denominados bvrr y bvrs por brucella virulence related) que forman parte de un sistema regulador de dos componentes. Este sistema present\u00f3 una homolog\u00eda superior al 70% con los sistemas chvi\/exos de rhizobium meliloti y chvi\/chvg de agrobacterium tumefaciens(bacterias filogen\u00e9ticamente proximas a brucella), implicados en la virulencia y el establecimiento de la relaci\u00f3n de la bacteria con celulas eucariotas. El an\u00e1lisis de los componentes de la ec mostr\u00f3 que los mutantes no expresaban la prote\u00edna omp25, una de las proteinas principales de la membrana externa. Dicha proteina ha sido recientemente incluida en una familia de la que varios de sus miembros est\u00e1n relacionados con la invasi\u00f3n celular y virulencia.
