El gradiente de protones creado es utilizado por la atpasa para la s\u00edntesis de atp. el objetivo del trabajo ha sido profundizar en el conocimiento del proceso de expulsi\u00f3n del prot\u00f3n al medio externo, as\u00ed como en la localizaci\u00f3n de los cationes divalentes que est\u00e1n fijados a la membrana purpura, los cuales son necesarios para un transporte correcta y eficaz.  para el estudio se construyeron los mutantes extracelulares e9q,e74q, e194q, e204q, e9q+e74q+e194q+e204q (4glu) y los citoplasm\u00e1ticos d36n+d38n y d102n+d104n.  las titraciones de los mutantes en medio \u00e1cido nos indican que el pka del asp\u00e1rtico 85, contrai\u00f3n de la base de schiff, est\u00e1 aumentado en d102n+d104n, e194q, e204q y 4glu en agua. Las diferencias con los resultados encontrados en 150 mm kc1 sugieren que estos grupos est\u00e1n involucrados en la uni\u00f3n de cationes. Por otro lado, el mutante 4glu presenta un equilibrio entre dos formas, forma roja y forma p\u00farpura, ya que las mutaciones han producido un cambio en el entorno del asp\u00e1tico 85, el cual puede ser restaurado por la adici\u00f3n de cantidades elevadas de i\u00f3n cloruro. Los estudios de dsc, asi como la reacci\u00f3n de la hidroxilamina indican que las mutaciones de los glut\u00e1micos 194 y 204 provocan una descompactaci\u00f3n de las h\u00e9lices, una menor estabilidad de la proteina, y un mayor acceso de la base de schiff al medio. Funcionalmente, el mutante e194q y 4glu parecen tener una producci\u00f3n de intermediario m disminuida, y los estadios por ftir indican que el pka del glut\u00e1mico glu 194
