Jose Calzada Garcia <\/strong><\/h2>\n<p>En la presente tesis doctoral, se plante\u00f3 el estudio estructural y funcional de la integrina plaquetaria aiibb3, que funciona como receptor de fibrin\u00f3geno y otras proteinas adhesivas. Este estudio se llev\u00f3 a cabo fundamentalmente con t\u00e9cnicas inmunoqu\u00edmicas, mediante las cuales, utilizando una colecci\u00f3n de anticuerpos monoclonales dirigidos a los subunidades de la integrina y cuyos ep\u00edtopos se han determinado en este trabajo, se han podido hacer experimentos que han permitido valorar la cantidad de receptor en superficie y la cantidad de receptor total, tanto en plaqueta humana como en plaqueta de otras especies animales de inter\u00e9s para un estudio comparado de dicho receptor, ya que estas especies son utilizadas como modelos experimentales de lesi\u00f3n vascular en cl\u00ednica. Tambi\u00e9n se ha hecho un estudio comparado del receptor de fibrin\u00f3geno(aiibb3) con el receptor de vitronectina (avb3) en celula endotelial humana, siendo tambien de gran interes, ya que ambos receptores tienen en comun la subunidad b3, y la subunidad aiib tiene una homolog\u00eda de un 74% con la subunidad av. Este estudio ha demostrado que dicha subunidad confiere a la subunidad b3 una conformaci\u00f3n distinta a la que adopta cuando est\u00e1 formando heterod\u00edmero con av en lugar de con aiib.  los estudios inmunoqu\u00edmicos de este receptor de fibrin\u00f3geno, tambi\u00e9n han permitido demostrar cambios estructurales en el mismo tras activaci\u00f3n, asi como la existencia de distintos estados de activaci\u00f3n agonista-dependientes, que presentan distintos cambios conformacionales.
