La adicion de glucosa a una suspension de celulas del mutante cif1\/tps1 provoca la acumulacion de hexosas-p, fructosa-1,6-p2 y triosas-p y la desaparicion del atp y el pi, lo que se interpreta como un desbalance entre las primeras etapas de la glicolisis y las siguientes. Hemos obtenido mutaciones supresoras extragenicas (sci1-1 y sc12-1) que permiten el permiten el crecimiento en glucosa del mutante cif1\/tps1. Ninguna de estas dos mutaciones restablecieron la sintesis de trehalosa. La mutacion sci1-1 presento defectos en la represion catabolica de ciertas enzimas, y probablemente suprime a cif1 por su disminucion en el transporte de glucosa. La clonacion de sci1 ha permitido su identificacion como cat3-snf4. La mutacion sci2-1 redujo al 50% la actividad fosforilante de glucosa, restableciendo de esta manera un patron silvestre de acumulacion de metabolitos tras la adicion de glucosa. El producto de la reaccion catalizada por cif1\/tps1, la trehalosa 6-p, ha resultado ser un potente inhibidor competitivo con glucosa y fructosa de las hexokinasas en s. Cerevisiae. La inhibicion ocurrio a concentraciones fisiologicas de trehalosa-6-p, segun las determinaciones realizadas por un nuevo metodo aqui descrito. A diferencia de lo que ocurre en s. Cerevisiae, la actividad hexokinasa de sch. Pombe resulto insensible a trehalosa-6-p y, consistentemente, la interrupcion del gen tps1+ en esta levadura no afecto al crecimiento en glucosa. Los resultados aqui presentados indican que la inhibicion de las hexokinasas por trehalosa-6-p constituye un mecanismo de regulacion importante de la glicolisis en s. Cerevisiae. En el presente trabajo tambien se ha demostrado el alelismo entre las mutaciones cif1 y fdp1 y se ha puesto de manifiesto que la trehalosa-6-p sintasa es necesaria para la
