Dado que el primer paso para que la insulina medie su accion es la union a su receptor celular (r1), y que este participa posteriormente en la transmision de la se\u00f1al hormonal, el objetivo de este trabajo ha sido estudiar el ri y asi determinar si la union de la insulina y los posteriores eventos que tienen lugar estan afectados por factores geneticos que pudiesen explicar esa propension a la ir que aparece en la enfermedad. Para alcanzar este objetivo hemos estudiado las caracteristicas cineticas de union de la insulina a su receptor, el destino que sigue la insulina unida a sus receptores de ebv-lonfocitos: y la capacidad de los ri para mediar la transmision de la se\u00f1al. Hemos encontrado: 1) que en el grupo de pacientes con dm hay una disminucion en la accion de la insulina sobre la autofosforilacion del ri.2) que en este grupo de pacientes con dm las caracteristicas de la interacion insulina-receptor (constantes de disociacion, cooperatividad, sensibilidad al ph) no muestran diferencias con las del grupo control. 3) que el destino que sigue la insulina unida a sus receptores de los ebv-linfocitos de los pacientes (disociacion, endocitosis del complejo insulina-receptor. Disociacion del complejo intracelular y degradacion de la insulina) es similar al grupo control. 4) que la expresion del gen de la dm es inducible por insulina en sujetos normales.
