{"id":84749,"date":"2018-03-10T00:09:08","date_gmt":"2018-03-10T00:09:08","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/estudio-de-la-resistencia-del-vih-1-a-antirretrovirales-en-un-modelo-in-vitro\/"},"modified":"2018-03-10T00:09:08","modified_gmt":"2018-03-10T00:09:08","slug":"estudio-de-la-resistencia-del-vih-1-a-antirretrovirales-en-un-modelo-in-vitro","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/ciencias-de-la-vida\/estudio-de-la-resistencia-del-vih-1-a-antirretrovirales-en-un-modelo-in-vitro\/","title":{"rendered":"Estudio de la resistencia del vih-1 a antirretrovirales en un modelo in vitro."},"content":{"rendered":"<h2>Tesis doctoral de <strong>  Iglesias De Ussel Perez M.dolores <\/strong><\/h2>\n<p>Nos planteamos el objeto de estudior, en un modelo in vitro, el proceso de aparici\u00f3n de variantes del vih-1 resistentes frente a distintas combinaciones de antirretrovirales.  para estudiar el proceso de emergencia de variantes resistentes hemos llevado a cabo la generaci\u00f3n y selecci\u00f3n de variantes mediante pases seriados en presencia de concentraciones crecientes de distintos antivirales (inhibidores de la retrotranscriptasa an\u00e1logos y no an\u00e1logos de nucle\u00f3sido, inhibidores de la proteasa y la hidroxiurea), tanto individualmente como en algunas de las posibles combinaciones dobles y triples.  las variantes obtenidos fueron caracterizados genot\u00edpicamente, por secuenciaci\u00f3n manual de las dos hebras del gen en estudio, tanto de la poblaci\u00f3n global como, en algunos casos, de la cuasiespecie. Tambi\u00e9n se llev\u00f3 a cabo la caracterizaci\u00f3n del fenotipo, mediante titulaci\u00f3n, determinaci\u00f3n de la susceptibilidad a los inhibidores y, en algunos casos, determinaci\u00f3n de la eficacia biol\u00f3gica.  hemos descrito un patr\u00f3n de desarrollo de resistencia en el que la emergencia de variantes resistentes aparece generalmente asociado a un retraso en la aparici\u00f3n del efecto citop\u00e1tico. Este proceso parece tener un componente estoc\u00e1stico, seguir distintos caminos evolutivos, aunque pr\u00f3ximos, y ser din\u00e1mico, por lo que una vez que se seleccionan variantes con una mutaci\u00f3n de resistencia el proceso contin\u00faa hacia la adquisici\u00f3n de m\u00e1s mutaciones de resistencia al mismo pero tambi\u00e9n a otro de los inhibidores empleados en la combinaci\u00f3n. As\u00ed, hemos podido comprobar que siempre que se utiliza la nevirapina emerge resistencia a este inhibidor y que, por el contrario, no hemos seleccionado ninguna variante resistente a estavudina.  tambi\u00e9n hemos seleccionado variantes con mutaciones de resistencia no descritas previamente in vitro, aunque s\u00ed en pacientes tratados.
