{"id":84832,"date":"2000-02-06T00:00:00","date_gmt":"2000-02-06T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/analisis-y-efectos-neuroqua%c2%admicos-del-acido-donoico\/"},"modified":"2000-02-06T00:00:00","modified_gmt":"2000-02-06T00:00:00","slug":"analisis-y-efectos-neuroqua%c2%admicos-del-acido-donoico","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/ciencias-de-la-vida\/analisis-y-efectos-neuroqua%c2%admicos-del-acido-donoico\/","title":{"rendered":"An\u00e1lisis y efectos neuroqu\u00edmicos del \u00e1cido donoico."},"content":{"rendered":"<h2>Tesis doctoral de <strong> Rafael Duran Barbosa <\/strong><\/h2>\n<p>El \u00e1cido domoico (dom) es un amino\u00e1cido tricarbox\u00edlico con una estructura relacionada con el \u00e1cido glut\u00e1mico y con el agonista glutamat\u00e9rgico \u00e1cido ka\u00ednico.  los objetivos del presente trabajo son:  1- desarrollar un m\u00e9todo y r\u00e1pido para el an\u00e1lisis y cuantificaci\u00f3n de los niveles de dom en muestras biol\u00f3gicas de moluscos, utilizando cromatograf\u00eda l\u00edquida (hplc) con detecci\u00f3n uv.  2- determianr los efectos del dom en sistemas in vitro, utilizando como modelo experimental la retina embrionaria de pollo.  3- estudiar los efectos del dom sobre los niveles de amino\u00e1cidos de inter\u00e9s neuroqu\u00edmico en muestras del tejido cerebral de rata adulta.  la utilizaci\u00f3n hplc isocr\u00e1tica en fase reversa en condiciones con una fase m\u00f3vil de actonitrilo\/agua al 15:85 y a ph 3,0 permite detectar niveles de dom inferiores a 1 ng por inyecci\u00f3n. En nuestras condiciones cromatogr\u00e1ficas no se han detectado niveles de dom t\u00f3xicos para el consumo humano en muestras de mejillones de la r\u00eda de vigo.  el dom produce mortalidad celular dependiente de la dosis pero no del tiempo en cultivos de c\u00e9lulas retinianas. Dicha mortalidad es dependiente de la presencia de calcio en el medio de cultivo, evitada por cnqx pero no por mk-801 y potenciada por la coadministraci\u00f3n de nmda.  el dom provoca un aumento considerable en los niveles de ip3 en las c\u00e9lulas retinianas, siendo dicho aumento evitado por cnqx pero no por mk-801. el ip3 generado por dom fue enteramente dependiente del calcio extracelular.  el aumento en la liberaci\u00f3n de glutamato inducido por dom es eliminado por efecto del cnqx, pero no por efecto del mk-801, mientras que el dom no provoca liberaci\u00f3n de aspartato en las mismas condiciones experimentales.  el dom provoca liberaci\u00f3n de gaba de manera parcialmente dependiente de calcio, siendo la liberaci\u00f3n independiente de calcio mayoritaria,
