Se desconoce si estas celulas poseen receptores para la vaso presina (avp) y la angiotensina ii (angii) y si poseen canales de ca2+ voltaje-dependientes (vocc). Los objetivos de la presente tesis doctoral son: 1)investigar la presencia de receptores de la avp y la avgii en hscs humanas en cultivo. 2)estudiar los efectos de dichas sustancias sobre contracci\u00f3n y proliferacion de las hscs.3)estudiar la expresion y de los vocc en hscs y su papel en la contraccion celular. Los experimentos de esta tesis se realizaron en hscs humanas mantenidas en cultivo. La presencia de receptores para la avp y la angii se demostr\u00f3 mediante la tecnicas de ligado. La movilizacion de ca2+ y la contraccion celular se estimaron en celulas cargadas con fura-2 y estudiadas en un microscopio de epifluorescencia acoplado a un sistema computarizado. La proliferacion celular se medi\u00f3 mediante la tecnica de la 3h-timidina y el contaje celular. Se ha demostrado que las hscs presentan receptores funcionales para la avp y la angii. La estimulaci\u00f3n de las hscs con dichas sustancias induce un marcado aumento dela [ca2+]i seguida de contraccion celular. Ademas, estas sustancias son mitog\u00e9nicas para las hscs. Las hscs humanas expresan numerosos vocc que median un aumento de la [ca2+]i, seguido de una intensa contraccion celular. Estos resultados confirman la capacidad contractil de las hscs e indican que la avp y la angii son dos potentes agonistas.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
