Cataliza la b-carboxilaci\u00f3n del fosfoenolpiruvato(pep) en presencia de hco3y mg2+ rindiendo oxaloacetato(oaa) y pi. Esta enzima ha sido muy estudiada en plantas c4 y cam, en las que juega un papel esencial en el metabolismo fotosintetico del carbono. La pepc est\u00e1 sometida la regulaci\u00f3n por fosforilaci\u00f3n reversible en un residuo de serina localizado en el extremo n-terminal de la proteina. La fosforilaci\u00f3n in vitro de la pepc induce el cambio en las propiedades cin\u00e9ticas y de regulaci\u00f3n de la enzima, aumentando su velocidad catal\u00edtica y afinidad por la glucosa-6-p, y dismuyendo la sensibilidad respecto a su efector negativo, el l-malato. Las enzimas responsables de la fosforilaci\u00f3n son las prote\u00ednas quinasas. Su identificaci\u00f3n, caracterizaci\u00f3n y la determinaci\u00f3n de su modo de activaci\u00f3n representa en gran parte el estudio de la se\u00f1alizaci\u00f3n en plantas. En este trabajo de tesis hemos caracterizado y purificado la(s)pepc-quinasa(s) de extractos crudos de hojas de sorgo iluminadas o mantenidas en oscuridad, as\u00ed como en fracciones purificadas,encontr\u00e1ndose actividad quinasa ca2+- dependiente y ca2+-independiente. Resultados obtenidos en este trabajo ponen en evidencia por primera vez que la regi\u00f3n c-terminal de la pepc est\u00e1 implicada en el reconocimiento y especificidad de la pepc-quinasa y aporten una prueba concluyente a favor de que la quinasa fisiol\u00f3gica que fosforila a la pepc sea la ca2+ -independiente. Adem\u00e1s nuestros resultados permiten afirmar que existen in vivo fen\u00f3menos de proteolisis implicados en la regulaci\u00f3n de la fosforilaci\u00f3n de la pepc. Por otra parte,hemos estudiado la pepc y pepc-quinasa de semillas. En semillas de cebada se muestra por primera que el h-7, un inhibidor de quinasas de tipo pka y pkc, es capaz de actuar in vivo inhibiendo la fosforilaci\u00f3n de la pepc.
