Para colonizar el sistema vascular el hongo debe ser capaz de atravesar las barreras estructurales de la planta, dentro de las que figuran la pared celular.La pectina es uno de los principales constituyentes de la pared de las c\u00e9lulas vegetales por lo que se piensa que la producci\u00f3n de enzimas pectinol\u00edticas participan en el mecanismo de patog\u00e9nesis de muchos microorganismos, entre ellos f. Oxysporum f.Sp. lycopersici.  el sistema pstinil\u00edtico de f. Oxysporum est\u00e1 constitu\u00eddo por al menos, seis pectinasas:pg1,pg2, pg3, pg5,pl1 y pl2. En este trabajo se aisla y caracteriza a la enzima pg3. Esta exopoligalacturonasa es secretada por f. Oxysporum f. Sp. Lycopersici durante el crecimiento del hongo en presencia de sustratos p\u00e9cticos como \u00fanica fuente de carbono. Una expresi\u00f3n similar tienen los genes pectinol\u00edticos pgx4 y pg5, tambi\u00e9n aislados durante esta tesis.  la presi\u00f3n de ambos genes no s\u00f3lo se ha observado in vitro sino tambi\u00e9n en plantas de tomate infectadas, lo que indica su participaci\u00f3n durante el proceso patog\u00e9nico de f. Oxysporum.Para aclara dicho papel, es necesario la inactivaci\u00f3n de los mismos y el an\u00e1lisis del comportamiento patot\u00edpico de los mutantes resultantes. F. Oxysporum hab\u00eda sido reportado como refractario a la integraci\u00f3n hom\u00f3loga de un dna ex\u00f3geno. La optimacizaci\u00f3n del m\u00e9todo de transformaci\u00f3n gen\u00e9tica, nos permiti\u00f3 concluir sobre el papel individual de pgx4 y pg5 en el proceso patog\u00e9nico de f. Oxysporum f.Sp. Lycopersici en su interaci\u00f3n con plantas de tomate.  el papel de las enzimas pectinol\u00edticas en el proceso de infecci\u00f3n de muchos fitopat\u00f3genos contin\u00faa siendo muy
