Recientemente se han desarrollado aines que muestran una actividad para cox-2 y carecen de los efectos secundarios atribuidos a la inhibici\u00f3n de cox-1. En esta tesis hemos analizado la expresi\u00f3n de cox-2 en dos modelos experimentales que cursan con una destacada respuesta inflamatoria: un modelo de artritis por ant\u00edgeno y otro de aterosclerosis por desecaci\u00f3n endotelial; estudiando adem\u00e1s la acci\u00f3n antiinflamatoria de un inhibidor selectivo de cox-2, el meloxicam. El incremento en el infiltrado de macr\u00f3fagos, observado tanto en las membranas sinoviales artriticas como en las femorales ateroscler\u00f3ticas, se correlacionaba con un aumento significativo en la expresi\u00f3n proteica y g\u00e9nica de cox-2; en cambio, los niveles proteicos de cox-1 sufrieron m\u00ednimas modificaciones. En ambos tejidos, cox-2 se localizan en las c\u00e9lulas inflamatorias as\u00ed como en las residentes. La administraci\u00f3n pofil\u00e1ctica de meloxicam a conejos con artritis preven\u00eda la respuesta inflamatoria al reducir el exudado (163+3 vs. 272+23 l\u00edquido sinovial artr\u00edtico) y la infiltraci\u00f3n polimorfonuclear (46 + 8 x 10 c\u00e9lulas\/ml l\u00edquido sinovial vs. 88+12&#215;10 c\u00e9lulas\/ml), pero no era capaz de aminorar el reclutamiento de monocitos. Por otra parte, la inhibici\u00f3n en la s\u00edntesis de pgs mediada por meloxicam (concentraci\u00f3n de pge2 5 veces inferior a la detectada en el l\u00edquido sinovial de animales sin tratar), se acompa\u00f1aba de un descenso de un 60% en la expresi\u00f3n g\u00e9nica de cox-2, sugiriendo una posible regulaci\u00f3n de dicha enzima por su producto. El tratamiento de c\u00e9lulas sinoviales (css) con pge2 confirm\u00f3 dicha hip\u00f3tesis, pures induc\u00eda un incremento en los niveles del mrna de cox-2 que fue m\u00e1ximo entre las 3 y 6h (1.6 veces respecto al basal con la dosis de 10 -6 m pge2).
