Esta proteina regula la proliferacion celular al inhibir la fosforilaci\u00f3n de prb por el complejo catal\u00edtico cdk4\/ciclina-d en la transici\u00f3n g1-s del ciclo celular. La deleci\u00f3n homocigota y la mutaci\u00f3n son mecanismos que provocan su inactivaci\u00f3n, aunque en algunas estirpes tumorales, como en el carcinoma colorrectal, no han sido descritos. La metilaci\u00f3n de la isla 5\u00c2\u00bf-cpg, situada en la regi\u00f3n promotora del gen, puede constituir un mecanismo alternativo de inactivaci\u00f3n. Analizamos la metilaci\u00f3n del gen p16\/cdkn2a mediante la tecnica de msp(metilaci\u00f3n especifica de pcr) en 86 tumores colerrectales, as\u00ed como 17 muestras de mucosa normal emparejadas con sus respectivas muestras tumorales. Ninguna de las muestras de mucosa normal mostr\u00f3 metilacion mientras que 18\/86 (20,9%, ci 95% 12,9%-31%) de las muestras tumorales mostraron metilaci\u00f3n del gen. Se observ\u00f3 una significativa correlaci\u00f3n entre la metilaci\u00f3n g\u00e9nica y la ausencia de expresi\u00f3n, analizada por inmunohistoqu\u00edmica, de la proteina p16 (p&lt;0,001). Adem\u00e1s, se analiz\u00f3 la posible relaci\u00f3n entre el estado de metilaci\u00f3n del gen p16 y diferentes factores clinico-patologicos definidos en el carcinoma colorrectal. No se encontr\u00f3 asociaci\u00f3n del estado de metilaci\u00f3n con la edad, el sexo, la localizaci\u00f3n, el estadio tumoral o la ploid\u00eda. Sin embargo, no puede descartarse una asociaci\u00f3n entre los tumores con metilaci\u00f3n y una mayor fase s del ciclo celular (p=0,088).
