Matilde Ram\u00edrez Alvarado <\/strong><\/h2>\n<p>La toxina del s\u00edndrome del shock t\u00f3xico (tsst-1) puede inducir la expresi\u00f3n del factor de necrosis tumoral-alfa (tnf-alfa) en c\u00e9lulas t y monocitos a trav\u00e9s de diferentes v\u00edas de se\u00f1alizaci\u00f3n. En esta tesis se muestran experimentos en los que se han estimulado c\u00e9lulas mononucleares de sangre perif\u00e9rica con tsst-1 y se ha encontrado por inmunohistoqu\u00edmica que las c\u00e9lulas encargadas de sintetizar tnf-alfa son los linfocitos. La expresi\u00f3n del tnf-alfa puede ser inhibida tanto como por wortmanina como por el ly294002, dos inhibidores de la fosfatidilinositol 3-quinasa (pi 3-k), adem\u00e1s, el efecto inhibidor no es transcripcional porque el tratamiento de las c\u00e9lulas con wortmanina o con ly294002 no cambia los niveles del mrna del tnf-alfa. el marcaje metab\u00f3lico y porterios inmunoprecipitaci\u00f3n del tnf-alfa intracelular mostr\u00f3 una disminuci\u00f3n de la s\u00edntesis del tnf-alfa en la c\u00e9lulas tratadas con los inhibidores de pi 3k.
